Cell Engineering with Functional Poly(oxanorbornene) Block Copolymers.

ABSTRACT: Cell-based therapies are gaining prominence in treating a wide variety of diseases and using synthetic polymers to manipulate these cells provides an opportunity to impart function that could not be achieved using solely genetic means. Herein, we describe the utility of functional block copolymers synthesized by ring-opening metathesis polymerization (ROMP) that can insert directly into the cell membrane via the incorporation of long alkyl chains into a short polymer block leading to non-covalent, hydrophobic interactions with the lipid bilayer. Furthermore, we demonstrate that these polymers can be imbued with advanced functionalities. A photosensitizer was incorporated into these polymers to enable spatially controlled cell death by the localized generation of ¹ O₂ at the cell surface in response to red-light irradiation. In a broader context, we believe our polymer insertion strategy could be used as a general methodology to impart functionality onto cell-surfaces.

Synthesis and Micellization Behavior of Amphiphilic Block Copolymers of Poly(N-vinyl Pyrrolidone) and Poly(Benzyl Methacrylate): Block versus Statistical Copolymers.

Project description:Block copolymers of N-vinyl pyrrolidone (NVP) and benzyl methacrylate (BzMA), PNVP-b-PBzMA, were prepared by RAFT polymerization techniques and sequential addition of monomers. The copolymers were characterized by Size Exclusion Chromatography (SEC) and NMR spectroscopy. Differential Scanning Calorimetry (DSC), Thermogravimetric Analysis (TGA) and Differential Thermogravimetry (DTG) were employed to study the thermal properties of these copolymers. The micellization behavior in THF, which is a selective solvent for the PBzMA blocks, was examined. For comparison the self-assembly properties of the corresponding statistical copolymers, PNVP-stat-PBzMA, were studied. In addition, the association behavior in aqueous solutions was analyzed for the block copolymers, PNVP-b-PBzMA. In this case, the solvent is selective for the PNVP blocks. Dilute solution viscometry, static (SLS) and dynamic light scattering (DLS) were employed as the tools to investigate the micellar assemblies. The efficient encapsulation of the hydrophobic curcumin within the micellar core of the supramolecular structures in aqueous solutions was demonstrated by UV-Vis spectroscopy and DLS measurements.

Synthesis and Characterization of Poly(ethylene furanoate)/Poly(ε-caprolactone) Block Copolymers.

Project description:Biobased poly(ethylene furanoate) (PEF)/poly(ε-caprolactone) (PCL) block copolymers have been synthesized using ring opening polymerization (ROP) of ε-caprolactone (ε-CL) in the presence of PEF in different mass ratios. An increase in intrinsic viscosity is observed for the block copolymers with higher ε-CL content due to the extension of their macromolecular chain. Matrix-assisted laser desorption ionization time-of-flight mass spectrometry (MS) was employed to understand the composition and structure of the produced block copolymers. The MS analysis helped to confirm the formation of PEF-PCL copolymers in all cases. Furthermore, tandem mass spectrometry experiments were performed to analyze the intrinsic fragmentation mechanism of neat PEF and PCL (both linear and cyclic) and confirm the block structure and end-groups. Finally, nuclear magnetic resonance results confirmed the composition and microstructure of the block copolymers. The synthesized PEF-PCL block copolymers can be used as a replacement for petroleum derived plastics, especially in the field of food packaging.

Synthetic Approaches for Poly(Phenylene) Block Copolymers via Nickel Coupling Reaction for Fuel Cell Applications.

Project description:Several methods to synthesize poly(phenylene) block copolymers through the nickel coupling reaction were attempted to reduce the use of expensive nickel catalysts in polymerization. The model reaction for poly(phenylene) having different types of dichlorobenzene derivative monomers illustrated the potential use of cost-effective catalysts, such as NiBr2 and NiCl2, as alternatives to more expensive catalysts (e.g., bis(1,5-cyclooctadiene)nickel(0) (Ni(COD)2)). By catalyzing the polymerization of multi-block poly(phenylene) with NiBr2 and NiCl2, random copolymers with similar molecular weights could be prepared. However, these catalysts did not result in a high-molecular-weight polymer, limiting their wide scale application. Further, the amount of Ni(COD)2 could be reduced in this study by approximately 50% to synthesize poly(phenylene) multi-block copolymers, representing significant cost savings. Gel permeation chromatography and nuclear magnetic resonance results showed that the degree of polymerization and ion exchange capacity of the copolymers were almost the same as those achieved through conventional polymerization using 2.5 times as much Ni(COD)2. The flexible quaternized membrane showed higher chloride ion conductivity than commercial Fumatech membranes with comparable water uptake and promising chemical stability.

Functional Polyglycidol-Based Block Copolymers for DNA Complexation.

Project description:Gene therapy is an attractive therapeutic method for the treatment of genetic disorders for which the efficient delivery of nucleic acids into a target cell is critical. The present study is aimed at evaluating the potential of copolymers based on linear polyglycidol to act as carriers of nucleic acids. Functional copolymers with linear polyglycidol as a non-ionic hydrophilic block and a second block bearing amine hydrochloride pendant groups were prepared using previously synthesized poly(allyl glycidyl ether)-b-polyglycidol block copolymers as precursors. The amine functionalities were introduced via highly efficient radical addition of 2-aminoethanethiol hydrochloride to the alkene side groups. The modified copolymers formed loose aggregates with strongly positive surface charge in aqueous media, stabilized by the presence of dodecyl residues at the end of the copolymer structures and the hydrogen-bonding interactions in polyglycidol segments. The copolymer aggregates were able to condense DNA into stable and compact nanosized polyplex particles through electrostatic interactions. The copolymers and the corresponding polyplexes showed low to moderate cytotoxicity on a panel of human cancer cell lines. The cell internalization evaluation demonstrated the capability of the polyplexes to successfully deliver DNA into the cancer cells.

Cubosomes from hierarchical self-assembly of poly(ionic liquid) block copolymers.

Project description:Cubosomes are micro- and nanoparticles with a bicontinuous cubic two-phase structure, reported for the self-assembly of low molecular weight surfactants, for example, lipids, but rarely formed by polymers. These objects are characterized by a maximum continuous interface and high interface to volume ratio, which makes them promising candidates for efficient adsorbents and host-guest applications. Here we demonstrate self-assembly to nanoscale cuboidal particles with a bicontinuous cubic structure by amphiphilic poly(ionic liquid) diblock copolymers, poly(acrylic acid)-block-poly(4-vinylbenzyl)-3-butyl imidazolium bis(trifluoromethylsulfonyl)imide, in a mixture of tetrahydrofuran and water under optimized conditions. Structure determining parameters include polymer composition and concentration, temperature, and the variation of the solvent mixture. The formation of the cubosomes can be explained by the hierarchical interactions of the constituent components. The lattice structure of the block copolymers can be transferred to the shape of the particle as it is common for atomic and molecular faceted crystals.

Biocompatible, degradable thermoplastic polyurethane based on polycaprolactone-block-polytetrahydrofuran-block-polycaprolactone copolymers for soft tissue engineering.

Project description:Biodegradable synthetic polymers have been widely used as tissue engineering scaffold materials. Even though they have shown excellent biocompatibility, they have failed to resemble the low stiffness and high elasticity of soft tissues because of the presence of massive rigid ester bonds. Herein, we synthesized a new thermoplastic polyurethane elastomer (CTC-PU(BET)) using poly ester ether triblock copolymer (polycaprolactone-block-polytetrahydrofuran-block-polycaprolactone triblock copolymer, PCTC) as the soft segment, aliphatic diisocyanate (hexamethylene diisocyanate, HDI) as the hard segment, and degradable diol (bis(2-hydroxyethyl) terephthalate, BET) as the chain extender. PCTC inhibited crystallization and reduced the melting temperature of CTC-PU(BET), and BET dramatically enhanced the thermal decomposition and hydrolytic degradation rate when compared with conventional polyester-based biodegradable TPUs. The CTC-PU(BET) synthesized in this study possessed a low tensile modulus and tensile strength of 2.2 MPa and 1.3 MPa, respectively, and an elongation-at-break over 700%. Meanwhile, it maintained a 95.3% recovery rate and 90% resilience over ten cycles of loading and unloading. In addition, the TPU could be electrospun into both random and aligned fibrous scaffolds consisting of major microfibers and nanobranches. 3T3 fibroblast cell culture confirmed that these scaffolds outperformed the conventional biodegradable TPU scaffolds in terms of substrate-cellular interactions and cell proliferation. Considering the advantages of this TPU, such as ease of synthesis, low cost, low stiffness, high elasticity, controllable degradation rate, ease of processability, and excellent biocompatibility, it has great prospects to be used as a tissue engineering scaffold material for soft tissue regeneration.

Intracellular trafficking of polyamidoamine-poly(ethylene glycol) block copolymers in DNA delivery.

Project description:The delivery of nucleic acids has the potential to revolutionize medicine by allowing previously untreatable diseases to be clinically addressed. Viral delivery systems have shown immunogenicity and toxicity dangers, but synthetic vectors have lagged in transfection efficiency. Previously, we developed a modular, linear-dendritic block copolymer architecture with high gene transfection efficiency compared to commercial standards. This rationally designed system makes use of a cationic dendritic block to condense the anionic DNA and forms complexes with favorable endosomal escape properties. The linear block provides biocompatibility and protection from serum proteins, and can be functionalized with a targeting ligand. In this work, we quantitate performance of this system with respect to intracellular barriers to gene delivery using both high-throughput and traditional approaches. An image-based, high-throughput assay for endosomal escape is described and applied to the block copolymer system. Nuclear entry is demonstrated to be the most significant barrier to more efficient delivery and will be addressed in future versions of the system.

Nanoconfinement-Induced β-Phase Formation Inside Poly(vinylidene fluoride)-Based Block Copolymers.

Project description:The electroactive properties of poly(vinylidene fluoride) (PVDF) are a direct consequence of its crystalline phases. Although poorly understood, nanostructuring PVDF in confined geometries can drastically change its crystallization behavior. Therefore, we synthesized a variety of PVDF-based triblock copolymers to gain a better understanding of the melt crystallization and explore how crystallization is affected by the morphology and chemical nature of the amorphous block. Differential scanning calorimetry, small-/wide-angle X-ray scattering, and transmission electron microscopy gave us excellent insights into the morphology and the corresponding crystalline phases. We find that crystallization of PVDF inside spherical nanodomains occurs via a homogeneous nucleation mechanism leading to a large undercooling and the formation of the thermodynamically favorable ferroelectric β-phase. On the contrary, when confined crystallization occurs inside a lamellar morphology, or in the case of breakout crystallization, a heterogeneous nucleation process leads to the formation of the nonferroelectric α-phase. Furthermore, favorable melt interactions between both blocks induce crystallization into the polar γ-phase at moderate cooling rates.

Synthesis and Characterization of Block Copolymers of Poly(silylene diethynylbenzen) and Poly(silylene dipropargyl aryl ether).

Project description:Poly(silylene diethynylbenzene)-b-poly(silylene dipropargyloxy diphenyl propane) copolymer (ABA-A), poly(silylene diethynylbenzene)-b-poly(silylene dipropargyloxy diphenyl ether) copolymer (ABA-O), and a contrast poly(silylene diethynylbenzene) with equivalent polymerization degree were synthesized through Grignard reactions. The structures and properties of the copolymers were investigated via hydrogen nuclear magnetic resonance, Fourier transform infrared spectroscopy, Haake torque rheometer, differential scanning calorimetry, dynamic mechanical analysis, thermogravimetric analysis and mechanical tests. The results show that the block copolymers possess comprehensive properties, especially good processability and good mechanical properties. The processing windows of these copolymers are wider than 58 °C. The flexural strength of the cured ABA-A copolymer reaches as high as 40.2 MPa. The degradation temperatures at 5% weight loss (Td5) of the cured copolymers in nitrogen are all above 560 °C.

Poly(oxanorbornene)-Coated CdTe Quantum Dots as Antibacterial Agents.

Project description:In this study, synthetic mimics of antimicrobial peptides based on poly(oxanorbornene) molecules (or PONs) were used to coat CdTe quantum dots (QDs). These PONs-CdTe QDs were investigated for their activity against Escherichia coli, a bacterium with antibiotic resistant strains. At the same time, the antibacterial activity of the PONs-CdTe QDs was compared to the antibacterial activity of free PONs and free CdTe QDs. The observed antibacterial activity of the PONs-CdTe QDs was additive and concentration dependent. The conjugates had a significantly lower minimum inhibitory concentration (MIC) than the free PONs and QDs, particularly for PONs-CdTe QDs which contained PONs of high amine density. The maximum activity of PONs-CdTe QDs was not realized by conjugating PONs with the highest intrinsic antibacterial activity (i.e., the lowest MIC in solution as free PONs), indicating that the mechanism of action for free PONs and PONs-CdTe QDs is different. Equally important, conjugating PONs to CdTe QDs decreased their hemolytic activity against red blood cells compared to free PONs, lending to higher therapeutic indices against E. coli. This could potentially enable the use of higher, and therefore more effective, PONs-QDs concentrations when addressing bacterial contamination, without concerns of adverse impacts on mammalian cells and organisms.
