ScRNAseq of splenic dendritic cells across age

ABSTRACT: Splenocytes from 1-week (n=5), 3-week (n=4), 4-week (n=3) and 6-week (n=3) old Clec9a-Cre Rosa26-tdTom mice were depleted of B cells, Neutrophils, Erythrocytes and T cells and labelled with barcoded antibodies according to timepoint. DCs were sort-purified and sorted DCs from two timepoints were pooled (1-week + 3-week; 4-week + 6-week) and then loaded onto two separate reactions of a Chromium chip (10X Genomics). Gene expression and Cell surface protein libraries were prepared using the Chromium Next GEM Single Cell 3’ Reagent kit (10X Genomics) according to manufacturer’s instructions. Concentration and purity of the libraries was analyzed with an Agilent TapeStation. Libraries were multiplexed and sequenced in accordance with 10X Genomics recommended sequencing depth, using NextSeq1000 (Illumina).
