Project description:Purpose: The goal of this sequencing is to investigate alterations in gene expression that result from impaired retinoid signaling compared with control, and how the RA signaling controls spermatogonia differentiation Methods: THY1+ spermatogonia mRNA profiles of 4-day-old control and germ cell specific impaired retinoid signaling mice were generated by High-throughput sequencing Results: Gene ontology (GO) analysis of the genes at the top of the ranked genes indicated enrichment in genes associated with roles in reproduction, transcription and spermatogenesis. In total, we identified 1633 and 742 transcripts (Reads Per Kilobase of transcript per Million mapped reads (RPKM) > 1) that were significantly (p-value < 0.05, > 1.5-fold difference) down- and up-regulated, respectively, in the germ cell mutants compared with the controls. Most importantly, we found that the majority of transcripts of replication-dependent core histone genes, histone cluster 1 (Hist1) were downregulated in germ cell mutants. RNA sequencing of aortic valves in CTRL, ECKO, SMKO and DKO mice

Project description:RNA sequencing was performed to compare the transcriptome of aortic valves among 4 mouse models: fibulin-4 (Fbln4; EFEMP2 gene) knockout in endothelial cells (EC) using Tie2Cre (ECKO), Fbln4 knockout in smooth muscle cells (SMC) using SM22Cre (SMKO), EC and SMC Fbln4 double knockout (DKO), and Fbln4loxp/+ and Fbln4+/+ with SM22Cre and/or Tie2Cre used as CTRL. At 2 months old, DKO mice showed significantly thickening aortic valve leaflets than other mouse models. Therefore, this experiment was used to investigate the function of Fbln4 in aortic valve development and Fbln4 role in EC. RNAseq analysis indicates differences in gene expression in aortic valves of DKO mice compared to other models, that is enriched with EndMT process and fibrosis progression.

GFRα1+, NGN3+ and KIT+ spermatogonia of adult mouse testes

Project description:In adult mouse testis, undifferentiated spermatogonia include GFRα1+ and NGN3+ populations. The former mainly act as self-renewing stem cells that support steady-state spermatogenesis. Whilst, the latter are primed for differentiation, but retain the potential of self-renewal and contribute to regeneration and post-transplantation colony formation (Nakagawa et al. science .2010; Hara et al. cell stem cell. 2014). And, NGN3+ cells can differentiate to KIT+ cells, which are also mentioned differentiated spermatogonia, by retinoic acid signal. The gene expression profiles of GFRα1+, NGN3+ and KIT+ spermatogonia, as well as the whole testis samples, were investigated here by microarray (Ikami et al. development. 2015).
