Project description:Caryopses of barley (Hordeum vulgare), like all other cereal seeds, are complex sink organs optimized for storage starch accumulation and embryo development. Their development from early stages after pollination to late stages of seed ripening has been studied in great detail. However, information on the caryopses’ diurnal adaptation to changes in light, temperature and alterations in phloem-supplied carbon and nitrogen remained unknown. In this study, we applied the 22K Barley1 GeneChip microarray to investigate diurnal gene regulation events of barley caryopses at 11 to 12 days post anthesis.

Pooled caryopses (form 3 plants) from the respective time points were used for RNA extraction and hybridization on Affymetrix microarrays (Barley1 GeneChip; GEO accsession GPL1340). Biological replicates were performed.

Project description:A large-scale parallel expression analysis was conducted to elucidate Mla-specified responses to powdery mildew infection using 22K Barley1 GeneChip probe arrays. Our goal was to identify genes differentially expressed in incompatible (resistant) vs. compatible (susceptible) and Mla-specified Rar1-dependent vs. -independent interactions. Project description:We describe here a new micro-proteomics workflow for the analysis of individual barley anthers. The workflow detects more than 4000 proteins from small amounts of material representing single or paired anther samples, covering a dynamic range of protein expression levels across five orders of magnitude. We consistently identified ~2800 proteins in a single 0.6mm anther and ~4000 proteins in a paired-anther sample. This demonstrates an important technological advance in plant proteomics, considering the limited amount of starting material. We employed our micro-proteomics workflow to investigate the proteome of the developing barley anther containing male gametes in the early stages of meiosis. We successfully identified several low abundant meiosis-related proteins, proving that our approach is highly sensitive and consequently powerful. To date, no work has been published previously on precisely staged individual plant anthers.

Project description:In the current project, we aimed to discover how the barley anther proteome changes, both quantitatively and qualitatively, during meiotic prophase I when the important process of homologous recombination is taking place. Taking advantage of a recently published barley anther transcriptomic dataset (BAnTr) and a newly developed sensitive mass spectrometry-based approach to analyse barley anther proteome, we have conducted high-resolution mass spectrometry analysis of barley anthers, collected at 6 time points and representing their development from pre-meiosis to metaphase. Each time point was carefully staged using immunocytology. We identified >6400 proteins, including 82 known and putative meiotic proteins, and were able to quantify dynamic variation in the proteome during different stages of barley anther development.

Project description:Affymetrix Barley1 GeneChip microarray was used to study the transcriptome of two fast neutron barley mutants FN362 and FN363 allelic to characterized necrotic leaf spot 3 (nec3) mutants. PCR screening of the 21 genes at least two-fold down-regulated in mutants did not show any deletions in the genome of mutants and, thus, failed to identify a gene responsible for the nec3 mutant phenotype suggesting that either probe set for the NEC3 gene is not on the Barley1 GeneChip, or that the expression of wt NEC3 is confined to specific developmental stage or tissue type.
