Microarray data of benzo[a]pyrene-treated WHCO1 vs EPC-2 cells

ABSTRACT: The aim of this project was to investigate the differential effects of benzo[a]pyrene (BaP) treatment on global gene transcription, comparing normal (EPC-2) and cancer (WHCO1) cell lines from the oesophagus, in order to further our understanding of the molecular mechanisms of BaP-induced development and maintenance of oesophageal squamous cell carcinoma.

Project description:Lack of change in microRNA expression in adult mouse liver following treatment with benzo(a)pyrene (BaP), as detected using Agilent miRNA arrays. We have investigated the effect of exposure to 150 mg/kg benzo(a)pyrene (BaP) for 3 days on mRNA and miRNA expression levels in adult mouse liver. We used Agilent miRNA array platforms to assess effects of BaP exposure on miRNA expression levels. Adult male mice were exposed to 150 mg/kg benzo(a)pyrene (BaP) or solvent for 3 days and sampled 4 hours after the last dose. MicroRNA expression levels in adult mouse liver were measured using Exiqon miRNA arrays. Our results indicate a distinct lack of effect of BaP of miRNA expression, despite widespread changes in mRNA levels (measured using Agilent arrays). Proteomic analysis on the protective effect of eriodictyol on benzo(a)pyrene-induced Caco-2 cytotoxicity

Project description:The present study was aimed to evaluate the protective effects of six different polyphenols against benzo(a)pyrene (BaP)-induced cytotoxicity in Caco-2 cells. The results shows that treatment of quinic acid, ferulic acid, homovanillic acid and BaP decreased the cell viability, whereas only co-treatment of 20 µM eriodictyol and naringenin reduced BaP-induced cytotoxicity, including cell apoptosis, cell cycle change and oxidative stress. Moreover, all results show that the inhibitory effect of eriodictyol was better than that of naringenin. Further, the potential protective mechanism of eriodictyol against BaP-induced toxicity was investigated by proteomics. We found that the genetic information processing pathway involves in 80 differentially expressed proteins (DEPs) showed the highest proportion and number of total proteins. Finally, eriodictyol inhibited BaP-induced cytotoxicity by regulating the expression of key proteins such as RPA2, SNRPA, RAD23B, NUP155 and AARS in transcription and translation. Overall, our results provide a new perspective for polyphenols inhibiting BaP-induced carcinogenesis.
