Genomic profiles of tremolite-induced malignant peritoneal mesotheliomas developed in rats


ABSTRACT: Intraperitoneal injection of tremolite initiates mesothelial injury and chronic inflammation that causes the development of malignant mesothelioma (MM). We performed high-resolution comparative genomic hybridization to identify characteristics in the genomic profiles of tremolite-induced MMs in rats. Overall design: The carcinogenesis protocol was performed by intraperitoneal injection of 10 mg of tremolite in F1 hybrid (crossing Brown-Norway and Fischer344) rats. Genomes of 8 malignant peritoneal mesothelioma were profiled on Agilent 180K rat genome CGH microarrays.

The onset of malignant mesothelioma (MM) is linked to exposure to asbestos fibers. Asbestos fibers are classified as serpentine (chrysotile) or amphibole, which includes the crocidolite, amosite, anthophyllite, tremolite, and actinolite types.
