Effect of BRCA1 overexpression on genetic expressions of cervical cancer based on next generation sequencing

ABSTRACT: The cells were cultured in DMEM medium supplemented with 10% fetal bovine serum (Sangon, China) and 1% antibiotic-antimitotic solution (Gibco, USA). The SiHa cells were transfected using Lipofectamine 3000 reagent following the manufacturer’s protocol and incubated for 48h for further analysis.The gene expression profiles were analyzed of SiHa cells transfected with GV230-BRCA1 or GV230 plasmid to assess effect of BRCA1 overexpression on genetic expressions of cervical cancer.

Project description:Primary liver cancer (PLC) is the third largest contributor to cancer mortality in the world. PLC is a heterogeneous disease that encompasses several biologically distinct subtypes including hepatocellular carcinoma (HCC), intrahepatic cholangiocarcinoma (ICC) and combined hepatocellular-cholangiocarcinoma (CHC). The cells were cultured in DMEM medium supplemented with 10% fetal bovine serum and 1% antibiotic-antimitotic solution. The HCC cell line LM3 and ICCA cell line RBE were transfected using Lipofectamine 3000 reagent following the manufacturer’s protocol and incubated for 48h for further analysis. The gene expression profiles were analyzed of LM3 and RBE cells transfected with si-FENDRR or si-NC to assess effect of FENDRR on genetic expressions of PLC.

NFkB signaling and ISGylation associated with BRCA1-mutated fallopian tube epithelium

Project description:Germline BRCA1 or BRCA2 mutations (mtBRCA1 and mtBRCA2) dramatically increase risk for high-grade serous ovarian cancer (HGSOC), the most commonly diagnosed histotype. Other risk factors for this cancer, which originates primarily in the distal fallopian tube epithelium (FTE), implicate ovulation. To test whether mtBRCA1 or mtBRCA2 FTE cells respond differently to peri-ovulatory follicular fluid (FF) exposure than control patient FTE, gene expression profiles from primary FTE cultures were compared at baseline, 24h after FF exposure, and 24h after FF replacement with culture medium. Hierarchical clustering revealed both FF exposure and BRCA mutation status affect gene expression, with BRCA1 mutation having the greatest impact. Analysis revealed increased NFκB and EGFR signaling at baseline, with increased interferon signaling after recovery from FF exposure in mtBRCA1 samples. Inhibition of EGFR signaling and ISGylation by increased BRCA1 expression was verified in an immortalized FTE cell line, OE-E6/E7, stably transfected with BRCA1. Suppression of ISG15 and ISGylated protein levels by BRCA1 expression was found to be mediated by decreased NFκB signaling and was transiently suppressed by FF exposure. This study demonstrates increased NFκB signaling associated with decreased BRCA1 expression resulting in increased ISG15 and ISGylation following FF exposure, which could represent potential targets for chemoprevention.

Research on mechanisms of miR-27a-3p on Cisplatin sensitivity of hepatocellular carcinoma based on next generation sequencing

Project description:Hepatocellular carcinoma (HCC) accounts for 75% of primary liver cancer, ranking seventh in the incidence rate of tumors worldwide, and second in the mortality rate of tumors. The 5-year survival rate after radical surgery is only 59%-68%, and due to the influence of proliferation, invasion, microvascular infiltration and inflammatory environment of hepatocellular carcinoma, Recurrence is still possible after surgical resection, local ablative therapy or liver transplantation. Systemic antitumor therapy can control disease progression and prolong patient survival, and a variety of kinase inhibitors are used in patients who cannot undergo surgery. However, due to the existence of acquired drug resistance to multikinase inhibitors, its effect is not good. Platinum-based FOLFOX4 is recommended, but its efficacy is limited by drug resistance. miR-27a-3p, located in the short arm of chromosome 19, has been found to be involved in inhibiting the activity, proliferation, angiogenesis, and invasion of liver cancer. Other studies have found that miR-27a-3p regulates the sensitivity of tumors (esophageal cancer and breast cancer) to Cisplatin by regulating the apoptosis of the Bcl-2 protein family. The cells were cultured in DMEM medium supplemented with 10% fetal bovine serum and 1% antibiotic-antimitotic solution. The HCC cell line Huh-7 was transfected using Lipofectamine 3000 reagent following the manufacturer’s protocol and incubated for 48h for further analysis. The gene expression profiles were analyzed of Huh-7 cells transfected with miR-27a-3p mimics or NC to assess effect of miR-27a-3p on Cisplatin sensitivity of HCC.

Pimary human keratinocyte knockdown with deltaNp63 specific siRNA

Project description:Human ΔNp63-specific siRNA was obtained from Invitrogen (Carlsbad, CA; sense: 5′-ACAAUGCCCAGACUCAAUU-3′; antisense: 5′-AAUUGAGUCUGGGCAUUGU-3′). Scrambled sequence siRNA for the negative control was purchased from Invitrogen. Transfections were performed using Lipofectamine RNAiMAX (Invitrogen) in Opti-MEM (GIBCO) at 40 nmol/L according to the manufacturer's instructions. The culture medium was replaced at 6 h after siRNA transfection. mRNA was extracted from NHEKs transfected with ΔNp63-specific or scramble sequence siRNA at 72 h after transfection. Microarray slides were scanned using a 3D-GENE human 25k (TORAY, Tokyo, Japan) and microarray images were automatically analyzed with AROSTM, version 4.0 (Operon Biotechnologies, Tokyo, Japan).

Expression data from mammary epithelial cells from BRCA1 mutation carriers and non BRCA1 mutation carriers

Project description:Female BRCA1 mutation carriers have a nearly 80% probability of developing breast cancer during their life-time. We hypothesized that the breast epithelium at risk in BRCA1 mutation carriers harbors mammary epithelial cells (MECs) with altered proliferation and differentiation properties. Microarray studies revealed that PMEC colonies from BRCA1 mutation carriers anticipate expression profiles found in BRCA1-related tumors, and that the EGFR pathway is upregulated in BRCA1 mutation carriers compared ton non BRCA1 mutation carriers.
