Project description:Organoid models provide powerful tools to study tissue biology and development in a dish. Here, we established first-time organoid models from early-postnatal (postnatal day 7) mouse molar and incisor, capable of differentiation toward ameloblast-like cells in vitro. To more in detail characterise organoids from mouse molar and incisor, bulk RNA-sequencing was performed on the following (1) early passage (passage 0) organoids from both tooth types grown in basal tooth organoid medium (TOM) with or without addition of exogenous epidermal growth factor (EGF); and (2) late passage (passage 5) organoids grown in TOM+EGF or differentiation medium (DM).

Project description:Cortical organoids generated from mESC from a Down Syndrome (DS) mouse model (Ts65Dn) were dissociated for single cell sequencing to understand whether differential gene expression in DS brains affects cell diversity and how the molecular events that control the development of these diverse types of neocortical neurons are affected by the trisomy. These results were generated in parallel to a single cell experiment in forebrain fetal tissue from WT and Ts65Dn mice.

Project description:A major technology for generating oocytes by in vitro gametogenesis involves generating ovary models called reconstituted ovaries (rOvaries). Widespread use of rOvaries depends on the reliable manufacturing of primordial germ cell like cells (PGCLCs) and fetal ovarian somatic cells (FOSCs). Using scRNA-seq, we identify that upon thaw, FOSCs are composed largely of Foxl2+ pre-granulosa cells, KRT19+ epithelial cells and Nr2f2+ mesenchymal cells which self-assemble together with PGCLCs into disc-like organoids containing multiple follicles embedded in NR2F2 stroma without an ovarian surface epithelium and no steroidogenic theca. The absence of an ovarian surface epithelium combined with the small size of GV-oocytes indicates that follicle production in the rOvary corresponds to first-wave folliculogenesis.

Project description:Objective: Despite organoid formation and characterization from different tissues have been extensively studied, pancreas remains one among the least investigated organs. Here we proposed a multifaced characterization of cell types present in human pancreatic organoids (hPO) and starting from their progenitor we described their maturation and their lineage relationships. Design: We performed unbiased transcriptome-wide scRNA-seq analysis on 3187 cells of human pancreatic organoids obtained from 3 healthy islet-depleted tissue. To support our results, we strengthened these findings with proteomic and qRT-PCR analyses performed during the long-term culture of hPO. Results: First, we described hPO focusing on their morphology and long-term culture molecular aspects and showing a well-defined epithelial organization supported by a genetic stability through all cell passages. Successively, based on single-cell RNA sequencing technology, we defined all cell types present in hPO confirming that our pancreatic organoid methodology is highly reproducible. Moreover, exploiting the latest bioinformatic tools and comparing our data with recently published ones we reshaped the conventional theory of univocal ductal nature of hPO. Finally, we defined a new strategy to sort a refined multipotent progenitor from hPO and we gave a new definition of pancreatic progenitor demonstrating its capacity to renew the hPO stem population while maintaining its heterogeneity. Conclusions: Taken together, our results give a new definition of human pancreatic organoids, which reveals the complex nature of these structures and provides a strong basis to pave the way for future study in the pancreatic field.

Project description:The cell-of-origin of high grade serous ovarian carcinoma (HGSOC) remains controversial, with fallopian tube epithelium (FTE) and ovarian surface epithelium (OSE) both considered candidates. Here, by using genetically engineered mouse models and organoids, we assessed the tumor-forming properties of FTE and OSE harboring the same oncogenic abnormalities. Combined RB family inactivation and Tp53 mutation in Pax8+ FTE caused Serous Tubal Intraepithelial Carcinoma (STIC), which metastasized rapidly to the ovarian surface. These events were recapitulated by orthotopic injection of mutant FTE organoids. Engineering the same genetic lesions into Lgr5+ OSE or OSE-derived organoids also caused metastatic HGSOC, although with longer latency and lower penetrance. FTE- and OSE-derived tumors had distinct transcriptomes, and comparative transcriptomics and genomics suggest that human HGSOC arises from both cell types. Finally, FTE- and OSE-derived organoids exhibited differential chemosensitivity. Our results comport with a dualistic origin for HGSOC and suggest that the cell-of-origin might influence therapeutic response.
