Large-scale RNAi screening uncovers new therapeutic targets in the human parasite Schistosoma mansoni

ABSTRACT: stk25 and tao genes came out of an RNAi screen in the parasitic flatworm Schistosoma mansoni, and these genes shared a similar knockdown phenotype. To test for a functional relationship, we compared transcriptomes of early knockdown worms for each gene, compared to mock controls.

Project description:RNAseq experiments to identify gene expression changes following gamma irradiation Adult male Schistosoma mansoni (strain: NMRI) were subjected to 200 Gy of gamma irradiation and cultured in vitro for 48hr hours. After 48hr germ line tissues were removed and RNA was extracted from remaining somatic tissues. Samples were processed for RNAseq using Illumina procedures.

Project description:Schistosoma mansoni is the causative agent of schistosomiasis, an endemic neglected tropical disease that affects the human population. Recently, newer versions of the genome and transcriptome have been published, with the Sanger Institute leading both the original publication of the genome as well as the further upgrade. However, little is known about the regulatory elements found in the genome, i.e. promoters. Identification of these elements is key for the understanding of mechanisms of regulation of gene expression in this parasitic helminth.
