Project description:Study to identify genes associated with NSCLC recurrence in patients not receiving adjuvant chemotherapy. Purpose: Recent clinical trials suggest improvement in survival with adjuvant chemotherapy in non-small cell lung cancer (NSCLC). This study's aim is to identify genes associated with NSCLC recurrence in patients not receiving adjuvant chemotherapy. Experimental design: Banked NSCLC tumors of patients who underwent resection of stage Ia-IIIb disease were identified. Patients were stratified into two groups: recurrent (R, n=11) or non-recurrent (NR, n=16) 2 years after surgery. Microarray profiling and Cox multivariate analysis were performed. Conclusion: Increased CYP3A5 gene expression correlates with NSCLC recurrence and promotes proliferation through mechanisms that may involve, in part, CYP3A5 epoxygenase activity. Sequencing of commercially-available pancreatic cancer cell lines reveals selective CYP3A5 overexpression among xenobiotic-metabolizing CYPs.

Project description:Purpose: To know the expression of all cytochrome P450 (CYP) enzymes and related drug metabolizing enzymes in pancreatic cancer models Methods: We obtained expression profiles of 8 commercially-available pancreatic cancer cell lines and one non-cancerous immortalized cell line by RNA-sequencing. The pancreatic cancer cells were compared among themselves and to the non-cancerous control cells. Results: We confirmed reports that CYP3A5 is highly overexpressed in pancreatic cancer models, and additionally report that known xenobiotic-metabolizing CYPs are lowly expressed in comparison.

TANK binding kinase 1 induces transcription factor BACH1 to regulate iron metabolism in pancreatic cancer cells

Project description:BTB and CNC homology 1 (BACH1) represses the expression of genes involved in the metabolism of iron, heme and reactive oxygen species and promotes metastasis of various cancers including pancreatic ductal adenocarcinoma (PDAC). Knockdown of Tank binding kinase 1 (TBK1) led to reductions of BACH1 mRNA and protein amounts in AsPC-1 human PDAC cells. Gene expression analysis of PDAC cells with knockdown of TBK1 or BACH1 suggested the involvement of TBK1 and BACH1 in the regulation of iron homeostasis. Ferritin was increased upon BACH1 knockdown in AsPC-1 cells. Cytometry analysis showed that AsPC-1 cells with BACH1 knockout or knockdown contained lower labile iron than control cells, suggesting that BACH1 increased labile iron by repressing the expression of ferritin genes.
