Quantitative analysis of standard diet and high fat diet-fed wild type and Sirt3 knockout mouse pancreatic islets transcriptomes.

ABSTRACT: To explore the underlying mechanism for the regulatory role of SIRT3 in pancreatic islets under standard and high fat diet feeding, we conducted RNA sequencing on the isolated islets from standard diet and high fat diet-fed wild type and pancreatic beta cell selective Sirt3 knockout mice (four groups in total). Three biological replicates were performed for each group.

Project description:Changes in the secretion profile of visceral-pancreatic white adipose tissue (pWAT) due to diet-induced obesity are partially responsible for increased beta cell replication, suggesting that a crosstalk between pWAT and beta cells may play a role in regulating beta cell plasticity. The molecular mechanisms underlying this cross-talk are still not fully understood. The aim of this study was to integrate transcriptomic, proteomic and metabolomic data to unravel the cross-talk between adipose tissue and pancreatic islets during evolution of obesity. Pancreatic islets from control lean and cafeteria diet fed obese rats were obtained. RNA was extracted and processed for further hybridization on Affymetrix microarrays (GeneChip Rat Genome 230 2.0 (Affymetrix, Santa Clara, CA)).

Project description:We report the application of next-generation sequencing technology for high-throughput profiling of H3K27ac and transcriptome analysis in pancreatic islets derived from C57Bl/6 mice fed a high-fat diet. We find genomic regions showing change in acetylation of histone H3K27 in response to long-term HFD feeding, which was significantly associated with differential gene expression. Furthermore, increased H3K27ac showed a distinctive genomic distribution surrounding proximal-promoter regions. This study provides a framework for the application of comprehensive chromatin profiling towards characterization of diverse mammalian cells under various environments.

Remodelling of the acetylproteome by SIRT3 manipulation fails to affect insulin secretion or beta-cell metabolism in the absence of over nutrition

Project description:From Peterson et. al. 2018: SIRT3 is an NAD+-dependent mitochondrial protein deacetylase purported to influence metabolism through post-translational modification of metabolic enzymes. Fuel-stimulated insulin secretion, which involves mitochondrial metabolism, could be susceptible to SIRT3-mediated effects. We used CRISPR/Cas9 technology to manipulate SIRT3 expression in b-cells, resulting in wide-spread, SIRT3dependent changes in acetylation of key metabolic enzymes, but with no appreciable changes in glucose- or pyruvate-stimulated insulin secretion, or in metabolomic profile during glucose stimulation. Moreover, these broad changes in the SIRT3-targeted acetylproteome did not affect responses to nutritional or ER stress conditions. We also studied mice with global SIRT3 knockout fed either standard chow (STD) or high- fat/high-sucrose (HFHS) diets. Only when chronically fed a HFHS diet do SIRT3 KO animals exhibit a modest reduction in insulin secretion. We conclude that broad changes in mitochondrial protein acetylation in response to manipulation of SIRT3 are not sufficient to cause changes in islet function or metabolism.
