Effects of Id1 depletion in TAMs on the genes expression of MC38 cells.

Project description:We report the application of high-throughput mRNA sequencing to explore the effect of Id1 depletion in TAMs on gene expression of MC38 cells. Different groups of TAMs were isolated from the subcutaneous tumors in Id1f/f or myeloid cell lineage speciﬁc Id1 deficient mice (Id1-Lyz-KO) . MC38 cells were then mixed with two groups of TAMs and be implanted in C57BL/6J mice. CD45- cells were then isolated from the subcutaneous tumors described above to do the transcriptome sequencing.

Next generation sequencing of a unique set of glycovariant MC38 mouse colorectal cancer cell lines generated with CRISPR-Cas9 and CRISPR-dCas-VPR technology

Project description:To investigate the effect of different tumor-associated glycans, mouse colorectal cancer cells (MC38 cell line) were genetically modified using CRISPR-Cas9 and CRISPR-dCas-VPR technology targeting different genes involved in glycosylation pathways. To study the effect of altered glycosylation at the transcriptional level, next generation sequencing was performed on the entire panel of glycovariant MC38 cell lines. The MC38 knockout cell lines created with the CRISPR-Cas9 technology are: MC38-MOCK#1, MC38-CMAS KO, MC38-COSMC KO, MC38-CMAS+COSMC KO. The MC38 overexpressing cell lines generated with the CRISPR-dCas9-VPR technology are: MC38-MOCK#2, MC38-FUT4, MC38-FUT9. Details on the cell lines can be found in the corresponding manuscript(s).

ALPN-202 induces a more potent anti-tumor T cell response in MC38/hPD-L1 tumors than PD-L1 blockade alone

Project description:Purpose: Use RNA-seq to characterize the anti-tumor immune response induced by ALPN-202 and compare to that of anti-PD-L1 treatment alone. Methods: mRNA was isolated from MC38/hPD-L1 tumors 72 hours after a single dose of ALPN-202 (n=4), anti-PD-L1 mAb (durvalumab) (n=4), or Fc control (n=4). Results: ALPN-202 treatment resulted in elevated expression of multiple T cell, NK cell, myeloid cell genes. Additionally, there was a strong increase in genes commonly associated with a proinflammatory response including cytokines, chemokines and surface markers. Conclusions: ALPN-202 treatment resulted in a strong anti-tumor immune response that was more potent than that generated by blockade of PD-L1 alone.
