Transcriptome dynamics of rice spikelets under heat stress treatment

Project description:Rice reproductive development is highly sensitive to high temperature stress. In rice flowering occurs over a period of at least 5 days. Heat stress alters the global gene expression dynamics in panicle especially during pollen development, anthesis and grain filling. Some of the rice genotypes like Nagina 22 show better spikelet fertility and grain filling compared to high yielding and popular rice cultivars like IR 64. We carried out microarray analysis of 8 days heat stressed panicles of Nagina22, heat and drought tolerant aus rice cultivar and IR64, a heat susceptible indica genotype along with unstressed samples of Nagina22 and IR64 so as to understand the transcriptome dynamics in these two genotypes under heat stress and to identify the genes important for governing heat stress tolerance in rice.

A histone deacetylase confers plant tolerance to heat stress by controlling protein lysine deacetylation and stress granule formation in rice

Project description:Elevated temperatures due to global warming seriously threaten crops production. Understanding molecular mechanisms of cellular responses to heat stress will help to improve crop tolerance to high temperature and yield. In this work, we show that deacetylation of non-histone proteins mediated by the rice cytoplasmic histone deacetylase HDA714 is required for plant tolerance to heat stress. HDA714 expression and protein accumulation are induced by heat stress. HDA714 loss-of-function affects specifically plant response to heat (42°C) while its over-expression enhances plant tolerance to the stress. Interestingly, heat stress led to decreases of overall protein lysine acetylation in rice plants, which depends on HDA714 function. HDA714-mediated deacetylation of metabolic enzymes stimulates glycolysis under heat stress. In addition, HDA714 protein is found within heat-induced stress granules (SGs), many identified rice SG proteins show lysine acetylation at normal temperature (25 °C), which is augmented in hda714 mutants. Finally, HDA714 interacts with and deacetylates several SG proteins and HDA714 loss-of-function impairs SG formation. Collectively, these results indicate that HDA714-mediated cellular protein lysine deacetylation responds to heat stress, affects metabolic activities, regulates SGs formation, and confers heat tolerance in rice plants.

Project description:In Arabidposis thaliana, the msh1 recA3 double mutant shows an extensive mitochondrial genome rearrangement and displays pronounced thermotolerance. We use microarray analysis to characterize genes and their response in the double mutant to a heat shock experiment in which plants were exposed to a temperature of 37 C for 2 hours. Wild type and double mutant plant were exposed to a temperature of 37 C for 2 hours and RNA samples were collected for the wild type and the double mutant before and after the heat shock.

Gel-free/label-free proteomic analysis of developing rice grains under heat stress

Project description:High temperature markedly reduces the yields and quality of rice grains. To identify the mechanisms underlying heat stress-induced responses in rice grains, proteomic technique was used. Khao Dawk Mali 105 rice grains at the milky, doughy, and mature stages of development after flowering were treated at 40 °C for 3 days. Aromatic compounds were decreased in rice grains under heat stress. The protein abundance involved in glycolysis and tricarboxylic acid cycle, including glyceraldehyde 3-phosphate dehydrogenase and citrate synthase, was changed in milky and doughy grains after heat treatment; however, no changes in mature grains. The abundance involved in amino acid metabolism was increased in doughy grains, but decreased in milky grains. In addition, the abundance involved in starch and sucrose metabolism, such as starch synthase, ADP-glucose pyrophosphorylase, granule-bound starch synthase, and alpha amylase, was decreased in milky grains, but increased in doughy grains. A number of redox homeostasis-related proteins, such as ascorbate peroxidase and peroxiredoxin, were increased in developing rice grains treated with heat stress. These results suggest that in response to heat stress, the abundance of numerous proteins involved in redox homeostasis and carbohydrate biosynthetic pathways may play a major role in the development of KDML105 rice grains.

Effect of knockdown of Musashi-1 in Group 3/4 medulloblastomas cells

Project description:Groups (Grp) 3 and 4 are aggressive molecular subgroups of medulloblastoma (MB), with high rates of leptomeningeal dissemination. To date, there is still a paucity of biomarkers for these subtypes of MBs. In this study, we investigated the clinical significance and biological functions of MSI1 in Grp3/Grp4-MBs. To that, we identified that the MSI1 knockdown sensitized D283 Med cells to cisplatin treatment by enhancing cell apoptosis and modulated cancer-related pathways. The results of the present demonstrate that MSI1 may play a role as biomarker for Grp3/Grp4-MBs. In addition, MSI1 knockdown combined with cisplatin may offer a potential strategy to be further explored in Grp3/Grp4-MBs.

We used microarrays to detail variation of the gene expression after cold treatment and screen more cold-response genes in rice.

Expression data from panicle in heat tolerant and heat susceptible rice under high temperature stress

Project description:To better understand the mechanisms that regulate the heat stress response in rice, we conducted a comparative analysis of transcriptome profile in panicles from two rice lines, heat-tolerant line 252 (HTL252) and heat-susceptible line 082 (HSL082) using rice Affymetrix GeneChip. In HTL252 panicles, 1538 differentially expressed genes (DEGs) genes with at least four-fold expression changes compared with the control under heat treatment were considered heat-responsive (HR). Of these DEGs, 522 genes were up-regulated while 1016 genes were repressed. Among DEGs in HSL082, 496 genes were induced and 1707 genes were repressed. Out of the 370 common DEGs found between HTL252 and HSL082, 129 genes were induced and 241 genes were repressed.

Thermal transcriptome of Synechocystis in response to gradual heat stress
