Multi-tissue single-cell atlas of murine atherosclerosis

ABSTRACT: Atherosclerosis is a chronic inflammatory disease in which both the innate and adaptive immune systems play a role. Our objective was to create a multi-tissue single-cell atlas of atherosclerosis using the hypercholesterolaemic Ldlr-/- Apob100/100 mouse model. We used mice fed with either a chow diet or a 3-month high fat diet to respectively represent an early state and a late state of the disease. We collected four tissues: epididymal white adipose tissue, perivascular adipose tissue, aorta, and spleen, and dissociated them into suspensions containing either all cells or enriched for CD45+ cells (aorta samples). The data has been created using 10X Genomics 5’ single-cell immuneprofiling technology.
