Project description:Gene expression profiles of Populus deltoides induced by fungal pathogenic Marssonina brunneain iduring time-course infection. We used Affymetrix poplar genome genechip microarrays to analyze the full transcript expression underlying time-course infection and identified significantly differently expressed genes during the infection process. 2-year-old seedlings of P. deltoides grown were innoculated with the suspension of the pathogenic spores throught leaves spraying. Collecting the samples at interval time of 2 days and one week. Affymetrix poplar genechip was hired to investigate the full transcripts changed of the poplar response to the pathogen attack during the time.

Project description:Populus euphratica is a natural population grown in semirid areas. The molecular response of the poplar to drought maintain to be elucided, especially at global genome level. We used Affymetrix poplar genome genechip microarrays to analyze the full transcript expression underlying different drought intensities and identified significantly differently expressed genes during this process. Uniformly developed seedlings of P. euphratica grown in gradually long-term drought throught water-withholding treatment. Affymetrix poplar genechip was hired to investigate the full transcripts changed of the poplar response to different drought intensity levels.

Identification of differentially regulated genes in Populus deltoides subjected to copper stress using 4.6K cDNA arrays

Project description:cDNA macroarray expression profiling was carried out in poplar roots in order to identify genes regulated in response to exposition to copper stress. For this purpose, plants of a Populus deltoides clone grown in a hydroponic system during four weeks were incubated in a nutrient solution (Hoagland's modiefied salt, ¼ strength) supplemented with copper (0 µM (control), 30 µM and 60 µM). Roots were sampled at 12 and 24 h after exposition in a time-course experiment.

Identification of rust-induced genes in poplar using Populus PICME 28K cDNA microarrays

Project description:Microarray expression profiling was carried out in poplar leaves upon infection with rust in order to identify genes expressed during tree defense response. For this purpose, we inoculated detached leaves of the interamerican hybrid poplar Populus trichocarpa x Populus deltoides 'Beaupré' grown in greenhouse with spores of avirulent strain 93ID6 of the pathogenic rust fungus Melampsora larici-populina (incompatible interaction, I48). Besides, we mock-inoculated 'Beaupré' leaves with water (control condition, T48). Detached leaves were maintained in vitro in controled conditions to allow fungal infection and colonization of plant tissue. Leaves were sampled 48 hours post-inoculation after that the fungus attempt to penetrate plant cells in mesophyll. Competitive hybridization between transcripts of incompatible interaction (I48) and control condition (T48) was done on Populus PICME 28K cDNA microarray. Project description:A microarray analysis of whole-genome gene expression in leaves was carried out in a (Populus trichocarpa X Populus deltoides) X Populus deltoides pseudo-backcross pedigree. Genetic variation in gene expression was quantified for 55,793 predicted gene models based on a single probe per gene. Resultant data contributed to the analysis of the genetic architecture of gene expression in leaves of Populus. Data include one biological replicate of 183 individuals segregating from a pseudo-backcross pedigree of (Populus trichocarpa X Populus deltoides) X Populus deltoides analyzed for gene expression (GE) in roots using one probe per gene for 55793 independent gene models (probes E_POPLARSxxxxxPxxxxx) and single feature sequence polymorphism (SFP) using one probe per gene for 12084 independent gene models (probes G_POPLARSxxxxxPxxxxx). GE and SFP probes were selected from 6-7 probes per gene previously tested in a pilot study of the two parent trees of the cross (Populus deltoides X Populus trichocarpa)
