A Brucella ovis HWE histidine kinase is important for detergent resistance.

Project description:Through a system-level analysis of non-essential two-component signal transduction systems (TCS) genes in Brucella ovis, we have discovered that deletion of an HWE histidine kinase, BOV_1602, leads to significant increase in resistance to SDS detergent and other anionic or zwitterionic detergents. To better understand this detergent resistance phenotype, we performed RNA-seq experiment using both WT and ∆BOV_1602 cultured on no stress (TASB) or TSAB supplemented with 0.004% SDS.

Gene expression profiles in rat lung following intratracheal instillation with C60 fullerene particles

Project description:Gene expression profiling of the rat lung following intratracheal instillation with C60 fullerene particles was employed to gain insights into these molecular events. Groups of nine-week-old male Wistar rats (n= 4-6 per group/ time point) were intratracheally instilled with C60 fullerene suspended in 0.4 ml distilled water including 0.8% Tween-80 as a single injection (0.1 mg, 0.2 mg (LD: low doses) and 1.0 mg (HD: high dose) C60 fullerene/ rat). Control groups received 0.8 % Tween-80 (vehicle control). After intratracheal instillation treatment, rats were housed within polycarbonate cages at a controlled temperature of 22 °C with a chow diet ad libitum, and dissected at 3 days, 1 week, 1 month, 3 month, and 6 month post-instillation. Right lungs of anesthetized rats were perfused with physiological saline, excised, and used for DNA microarray analysis.

Tween 20 inducible genes

Project description:Polyethylene glycol sorbitan monoacylates (Tween) are detergents of widespread use in plant sciences. We show them, notably Tween 20, to cause a rapid and complex change in transcript abundance which bears all characteristics of a PAMP / elicitor-induced defense response, and they do so at concentrations which cause no detectable deleterious effects on plant cellular integrity. The activity does not reside in the intact Tween molecule itself, but is caused by medium-chain fatty acids, notably lauric acid (LA), which are efficiently released from the Tween-backbone by the plant. The Tween / LA-response is independent of the jasmonate signalling system. Medium-chain fatty acids are thus novel elicitors/regulators of plant pathogen defense. The results also have several practical implications: (i) The use of Tweens and, as we show, several other detergents, as solvating/wetting agents on intact plants causes profound physiological changes which may mask actual effects of test compounds; (ii) Tweens by themselves can be regarded (and probably used) as economical, non-toxic, and safe-to-apply elicitors of inducible plant immunity against pathogens. Comparison of gene expression of WT and PerM mutant M. tuberculosis H37Rv grown in high and reduced magnesium

Project description:M. tuberculosis membrane protein PerM is required for persistence in chronic mouse infection. In vitro, the perM mutant requires increased magnesium for replication and survival, with elongation and lysis in low-magnesium media. To gain insight into the function of PerM, we compared the transcriptomes of wt and perM mutant Mtb grown in high (2000 M-NM-<M) and reduced (250 M-NM-<M) Mg2+ in three independent experiments. Transcriptome analysis of the perM mutant grown in reduced magnesium revealed upregulation of cell division and cell wall biosynthesis genes. Bacteria were grown for 5 days in SautonM-bM-^@M-^Ys media containing 250 or 2000 M-NM-<M MgCl2 and 0.05% Tween 80. Three independent experiments were performed.

Detergent-resistant membranes in Agrobacterium tumefaciens

Project description:We established a protocol for the biochemical fractionation of Agrobacterium tumefaciens membranes, and characterized the proteome of detergent-sensitive and detergent-resistant membrane fractions by mass spectrometry. We found that well-established microdomain marker proteins co-purify with proteins associated with transport or secretion processes in the detergent-resistant fraction, suggesting a functional micro-compartmentalization of the Gram-negative membrane. The presence of components of virulence-associated secretion systems in detergent-resistant membranes supports the involvement of bacterial microdomains in host-microbe interaction and microbial competition.
