Multi-omics of Sex-Based Differences in Nephrotoxic Serum Nephritis [scATAC-seq]

ABSTRACT: Sex-based differences have been observed in the incidence and clinical course of acute kidney injury as well as chronic kidney disease. Compared to males, female patients are more protected from kidney injury and disease progression. Prior studies have identified sexual dimorphism in kidney development and ischemic response, but the mechanisms by which inflammatory stimuli contribute to kidney sex based differences have not been fully elucidated. To examine these differences, we induced nephrotoxic serum (NTS) nephritis in female and male C57BL/6 mice of either 8-12 weeks or 18 months of age. Kidneys were harvested on Day 10. Nuclei were isolated and multiomics sequencing (matched snRNA-seq and snATAC-seq) was performed on individual nuclei.

PGE2 promotes recovery from established Nephrotoxic Serum Nephritis in mice.

Project description:We induced acute nephritis by single injection of sheep nephrotoxic serum in mice and then treated the mice with prostaglandin E2. We used microarrays to examine the global gene expressions during recovery from nephrotoxic nephritis by prostaglandin E2. Acute nephristis was induced in female mice by single injection of sheep Nephortoxc Serum (NTS), followed by prostaglandin E2 administration daily starting from day 2 after NTS administration. Mice were sacrificed on day 7 and RNAs were isolated from kidney. The RNA samples were subjected to mouse gene 1.0 ST array analysis.

The direct effect of mycophenolate mofetil on podocytes in nephrotoxic serum nephritis

Project description:The goal of our study was to characterize glomerulus and particularly podocyte biology during MMF treatment in an immune-triggered proteinuric glomerulopathy. Therefore, nephrotoxic serum nephritis was induced in three-week old wild-type mice. On day 3, half of the mice were treated with MMF (100 mg/kgBW/d p.o.) (NTS+MMF) for one week, the other half of animals with vehicle (NTS+veh). A further group without induction and treatment served as controls (C). On day 10, we performed proteomic analysis of glomeruli.

Project description:Studying the differences in glycosylation in B cells of normal mice and mice with an autoimmune disease (nephrotoxic nephritis). Differences in glycosylation in B cells of normal mice and mice with an autoimmune disease (nephrotoxic nephritis) were studied via gene expression analysis. RNA from mouse resting spleen B cells and B cells stimulated with antiCD40, CpG, and IL4 for 5 days, was isolated and prepared. One sample for each class was prepared. RNA was labeled and hybridized to the GLYCOv3 array. Resulting Gene expression patterns were analyzed.

RNAseq of kidneys from a mice model of chronic leptospirosis infection and induced a secondary nephrotoxic injury by adenine feeding

Project description:Background The mysterious chronic kidney disease is multifactorial causes. One of the risk factors is leptospirosis, a re-emerging infectious disease caused by Leptospira spp., for endemic leptospirosis tend to coincide with high-incidence regions of chronic kidney disease of unknown etiology. This study aims to investigate the role of leptospirosis as an emerging culprit in which chronic subclinical kidney injury, may predispose to progressive kidney disease when superimposed on secondary nephrotoxic injury. Methods Adenine-induced renal injury in chronic leptospira-infected C57/BL6 mice were studied for evaluating the renal function, histology and gene expression changes. We employed RNA sequencing-based renal transcriptome to investigate pathogenic pathways associated with secondary nephrotoxic injury in chronic kidney injury due to leptospirosis. Results The severity of kidney lesions was increased and the expression of immune/inflammation/fibrosis genes were significantly up-regulated in either low-dose (0.1%) and high-dose (0.2%) adenine-induced renal injury superimposed on infected mice. Whole renal transcriptome analysis reveals that the substantial amplification of the tremendous amount of expressed genes and fibrosis-related pathways was found in adenine-induced kidney injury add-on to leptospira-infected mice which correlated with kidney dysfunction and fibrosis compared to infection or adenine itself. A total of 41 differentially expressed genes associated with fibrosis were identified in infected mice fed with adenine, suggesting that these potential genes contributed to aggravated renal progression occurred in adenine-fed chronic leptospira-infected mice. Conclusions In summary, this study indicates that chronic subclinical kidney infection when exposed to different degree of secondary nephrotoxic injury may predispose to exacerbated and progressive chronic kidney disease.
