Project description:To investigate the effect of Lacticaseibacillus rhamnosus 0030 (LR) on nucleus accumbens gene expression in a mouse model of established diet-induced obesity, we treated high-fat diet fed male mice with 1x10^8 CFU LR using daily peroral gavage. Vehicle (here: PBS)-treated mice fed with a low-fat diet or high-fat diet served as controls. We then performed gene expression profiling analysis using data obtained from RNA-seq of those three groups (LFD vehicle, HFD vehicle, HFD LR).

Effect of beta3 adrenergic activation on liver gene expression profile in mice (C57Bl/6J) expressing hepatocyte PPARalpha or not

Project description:Pparα hepatocyte-specific knockout (Pparαhep-/-) mice were generated at INRAE’s rodent facility (Toulouse, France) by mating the floxed-Pparα mouse strain with C57BL/6J albumin-Cre transgenic mice to obtain albumin-Cre+/-Pparαflox/flox mice (LKO). Albumin-Cre-/-Pparαflox/flox (Pparαhep+/+) littermates were used as controls (LWT). Twelve week-old mice were transferred in a ventilated cabinet at the specific temperature of 30°C (thermoneutrality) 2 weeks before experiments. Mice were fed ad libitum or fasted at ZT0 and given beta3 adrenergic receptor agonist: CL316243 ((3 mg/kg body weight; Sigma Aldrich) = Beta3) or vehicle (0.5% carboxymethyl cellulose in sterilized water)=vehicle) by gavage at ZT10 and sacrificed at ZT16.

Effect of Sunitinib treatment withdrawal on Immunodeficient mice bearing MDA-MB231 tumors

Project description:Immunodeficient RAG1-/- mice bearing MDA-MB231-LUC+ tumors of 50-70 mm3 were daily administrated by gavage with vehicle (containing, 0.5% carboxymethylcellulose, 1.8 % NaCl, 0.4 % Tween-80, 0.9 %. benzyl alcohol, and ultrapure water adjusted to pH 6.0) or sunitinib (40mg/kg/day) for 30 days followed by treatment withdrawal for 3-4 weeks to allow the tumor regrowth until reaching the same volume as that of the vehicle treated group. Mice were sacrificed when tumor reached the volume of 350-400 mm3 and RNAs of two tumors from two different mice treated with sunitinib or vehicle were extracted.

Bisphenol A-induced renal metabolic disruption: Insights into MAPK-mediated steroid hormone dysregulation from transcriptomic and metabolomic analyses

Project description:After long-term exposure of BPA, human is probably suffering from invisible renal injury and metabolic disruption, while its linked intricate gene transcriptional mechanisms warrant more conclusive evidence. Comprehensive analysis utilizing transcriptomics and metabolomics were used to elucidate the transcriptional mechanisms regulating metabolic homeostasis involved in BPA-induced nephrotoxicity. Here, six-week-old male ICR (CD1) mice were randomly divided into vehicle group with 10 mice and BPA exposure group with 10 mice (five mice per cage). The mice in the exposure group received a daily gavage of BPA dissolved in an aqueous solution containing 0.1% ethanol at the dose of 50 μg/kg/d, which is the tolerable daily intake (TDI) dose of BPA. The mice in the vehicle group received the same dose of aqueous solution containing 0.1% ethanol. After 6-week exposure, the mice were euthanized and left kidney were tissues were frozen with liquid nitrogen and used for RNA transcriptome sequencing.

Global hepatic gene expression data from PPARa liver KO, PPARa liver WT, PPARaKO and WT male mice treated or not with Fenofibrate

Project description:Fenofibrate is a specific agonist of the nuclear receptor PPARa. To identify the gene expression under the strict dependence of hepatic PPARa activity, we generated a new mouse strain of PPARa-specific deletion in hepatocyte (albumin-Cre+/- Pparaflox/flox or LKO) and we compared them to total Ppara KO (KO), wild-type (WT) and liver WT (albumin-Cre-/- Pparaflox/flox or LWT) mice. We used microarrays to detail the global programme of gene expression in liver of Ppara LKO, LWT, Ppara KO and WT male mice. There are 36 liver samples, each from an individual mouse. The samples are from Ppara liver KO (LKO), Ppara KO (KO), wild-type (WT) and liver WT (LWT) male mice of 14 week-old from the same genetic background (C57Bl/6J) treated with Fenofibrate (100 mg/kg/day) or vehicle (aqueous solution of gum Arabic 3%) by daily gavage for 10 days. n= 4 mice for LKO, LWT and WT genotypes treated with vehicle; n=3 for KO mice treated with vehicle; n=5 mice for LWT, LKO and KO genotypes treated with fenofibrate; n=4 WT mice treated with fenofibrate. All mice were sacrified at ZT14.
