MicroRNA profiling to identify the biological pathways potentially associated with the actions of IVIg treatment


ABSTRACT: Purpose: Our study has demonstrated favorable effects of intravenous immunoglobin (IVIg) treatment on acute neurological status, motor function, anxiety level, and spatial learning ability after severe TBI in a controlled cortical impact (CCI) mouse model. We conducted high throughput small RNA-Seq to identify the biological pathways potentially associated with the actions of IVIg treatment. Methods: Differentially expressed (DE) miRNAs were identified using pairwise comparisons: a. TBI vs. sham, b. TBI-IVIg vs sham, and c. TBI vs TBI-IVIg (n =8 each treatment group), from brain samples and plasma samples collected 48 hours after TBI. Small RNAs were sequenced with 51 bp single-end reads using Illumina HiSeq 4000 and read quality was assessed using FastQC.
