Transcriptome of Ag-specific B cells upon stimulation with soluble or particulate Ag

ABSTRACT: Primary B cells from MD4 mice have transgenic B cell receptors that recognize hen egg lysozyme (HEL). Previous studies have shown that antigens presented in different formats induce qualitatively different signaling responses in B cells. Here, we compare the transcriptomes induced by the following stimuli: LPS (10 mg/mL), particulate HEL (1pM liposomes with reduced affinity HEL-3X protein conjugated to surface at epitope density 243), soluble HEL (1mg/mL), and co-stimulation with soluble HEL and anti-CD40 (100 ng/mL).

Transcription profiling by array of human A2780 ovarianc cancer cells and cisplatin resistant derivate cells treated with cisplatin or liposomal cisplatin

Project description:A2780 ovarian cancer cells and a cisplatin resistant derivate of A2780 cells, obtained from ECACC, UK, No. 93112519 [A2780] and No. 93112517 [A2780 cis] were seeded out in T-75 flasks at a density of 2x106 for A2780sens and 3x106 for A2780cis cells in 15 ml of medium and preincubated overnight. Medium was removed and 15 ml fresh medium (37M-0C) with different concentrations of cisplatin, liposomal cisplatin or empty liposomes were added and incubated for 72 h at 37M-0C in a 5% CO2 incubator. In case of A2780sens cells, 1.72 M-5M cisplatin (IC50 concentration) and in case of A2780cis cells 8.94 M-5M cisplatin (IC50 concentration) were added. Liposomal formulations contained equal cisplatin concentrations. Empty liposomes were added in the same concentration as the liposomal cisplatin, to analyze the impact of liposomal lipids (A2780sens: 0.80M-5mol lipid, A2780cis: 4.15 M-5mol lipid). After incubation, medium was removed and cells were washed thrice with 10 ml PBS. 1 ml RLT-buffer was added and cells lysates were stored at -80M-0C until RNA extraction.

Phenotypes of Th lineages generated by the commonly used activation with anti-CD3/CD28 antibodies differ from those generated by the physiological activation with the specific antigen

Project description:T-helper (Th) lineages have been generated in vitro by activating CD4 cells with anti-CD3/CD28 antibodies during polarization. Physiologically, however, the generation of Th lineages is by activation with the specific antigen presented by antigen-presenting cells (APC). Here, we used TCR-transgenic mice to compare the phenotypes of Th1, Th9 and Th17 lineages when generated by either one of the two activation modes. Lineage Th cells specific against hen egg lysozyme (HEL), were adoptively transferred into recipient mice transgenically expressing HEL in their lens. Remarkable differences were found between lineages of Th1, Th9, or Th17, generated by either one of the two modes in their capacities to migrate to and proliferate in the recipient spleen and, importantly, to induce inflammation in the recipient mouse eyes. Substantial differences were also observed between the lineage pairs in their transcript expression profiles of certain chemokines and chemokine receptors. Surprisingly, however, close similarities were observed between the transcript expression profiles of lineages of the three phenotypes, activated by the same mode. Furthermore, Th cell lineages generated by the two activation modes differed considerably in their pattern of gene expression, as monitored by microarray analysis, but exhibited commonality with lineages of other phenotypes generated by the same activation mode. This study thus shows that (i) Th lineages generated by activation with anti-CD3/CD28 antibodies differ from lineages generated by antigen/APC and (ii) the mode of activation determines to a large extent the expression profile of major transcripts NaM-CM-/ve CD4+ T cells purified from spleen and lymph node cells of 3A9 mice were activated and polarized toward Th1, Th9, and Th17 lineages, under either plate bound anti-CD3/anti-CD28 (PBAB) or APC presented HEL protein (HA).

Resolution of inflammation in imprinted fibroblast-like synoviocytes establishes a transcriptomic signature that promotes endothelial cell dysfunction.

Project description:Purpose: We analyzed RNA-seq from fibroblast-like synoviocytes obtained from TNF induced inflammatory arthritis mice. Our goal was to evaluate the gene expression signatures after the cessation of inflammatory conditions. Methods: We analyzed bulk RNA-seq from fibroblast-like synoviocytes from TNF induced inflammatory arthritis mice in diferent conditions: homeostasis (5% sucrose water for 3 weeks), inflammation (1mg/ml Doxycycline + 5% sucrose water for 3 weeks), resolution (1mg/ml Doxycycline + 5% sucrose water for 3 weeks and then stopped for next 3 weeks). Samples were sequenced to determine transcriptional changes and regulated processes. Results: paired-end reads were mapped to the mus musculus GRCm38.p3 genome assembly. RNA levels were normalized by RPKM and student's t-test. Conclusion: Despite the resolution of the inflammatory phase of arthritis, fibroblast-like synoviocytes showed a clear persistence of inflammatory transcriptomic expression. Our study highlighted that the persistent aggressive phenotype of fibroblast-like synoviocytes not only promotes inflammation and exacerbates joint damage but also triggers impaired endothelial cell functioning.
