ABSTRACT: E-liquid flavorings are one of the driving attractors for young electronic cigarette users. The added flavors are considered safe for oral ingestion, however long-term respiratory effects remain unknown and unassessed. This study aimed to systematically investigate the intrinsic toxicity of individual flavors in a submerged lung cell model and in aerosolized form having a more complex composition using an air-liquid interface (ALI) lung cell model. 16HBE14o- bronchial epithelial cells were exposed in submerged condition for 24 h to non-cytotoxic doses of cinnamaldehyde, eugenol, vanillin and diacetyl. Cells were exposed at ALI to aerosols of self-made e-liquids containing 70 µg mL-1 cinnamaldehyde, eugenol, or vanillin for 10 min with 24 h post-exposure. Transcriptomic changes were analyzed using the Clariom™ S assay in order to understand mechanistic toxicity due to flavoring exposure.
