Project description:Knee osteoarthritis contributes substantially to worldwide disability. Post-traumatic osteoarthritis (PTOA) develops secondary to a joint injury, such as ligament rupture, and there is increasing evidence suggesting a key role for inflammation in the etiology of PTOA and associated functional deficits. Colony stimulating factor 1 receptor (CSF1-R) has been implicated in the pathogenesis of musculoskeletal degeneration following anterior cruciate ligament (ACL) injury. We sought to assess the efficacy CSF1-R inhibition to mitigate muscle and joint pathology in a mouse model of PTOA. Four-month-old mice were randomized to receive a CSF1-R inhibitor and studied for 7 or 28 days after joint injury. Additionally, we profiled synovial fluid samples for CSF1-R from patients with injury to their ACL. Transcriptomic analysis of quadriceps muscle and articular cartilage in CSF1-R inhibitor-treated animals at 7 days after injury revealed elevated chondrocyte differentiation within articular cartilage and enhanced metabolic and contractile gene expression within skeletal muscle. At 28 days post-injury, CSF1-R inhibition attenuated PTOA severity and mitigated skeletal muscle atrophy. Patient synovial fluid CSF1-R levels correlated with matrix metalloproteinase 13, a prognostic marker and molecular effector of PTOA. Our findings support an opportunity for CSF1-R targeting to mitigate the severity of PTOA and muscle atrophy after joint injury.

Project description:Post traumatic osteoarthritis (PTOA) is a painful joint disease characterized by the degradation of bone, cartilage, and connective tissues. PTOA is initiated by trauma to joint-stabilizing tissues, such as the anterior cruciate ligament, medial meniscus or by intra-articular fractures. In humans, ~50% of joint injuries progress to chronic PTOA, while the rest spontaneously resolve. To understand molecular programs contributing to PTOA development, we examined injury-induced fluctuations in immune cell populations and transcriptional shifts by single-cell sequencing of synovial joints in PTOA -susceptible C57BL6/J (B6) and -resistant MRL/MpJ (MRL) mice. A potent myeloid driven anti-inflammatory response was discovered in MRL injured joints that significantly contrasted the pro-inflammatory signaling seen in B6 joints. We found Trem2+ macrophages classically described as M2 (CD206+Trem2+) to be enriched in the uninjured joints of MRL mice, and these populations were consistently elevated in this strain, post injury. To evaluate whether Trem2 is essential to macrophage-mediated homeostasis in the joint, we examined Trem2 deficient mice and determined that these mice develop spontaneous OA. Overall, these data suggest that the PTOA resistant MRL mouse strain displays enhanced capacity of clearing debris and apoptotic cells induced by inflammation after injury due to an increase in activated macrophages within the synovial tissue and joint space. Furthermore, we showed that Trem2 is essential for preserving cartilage homeostasis in B6, and we postulate that the elevated levels of Trem2+ joint-resident macrophages in MRL are in part responsible for the PTOA-resistant phenotype in this strain.

Project description:Injuries to the anterior cruciate ligament (ACL) often result in post-traumatic osteoarthritis (PTOA). PTOA accounts for ~12% of all osteoarthritis (OA) cases, yet the mechanisms contributing to OA after joint injury are not well understood. To better understand the molecular mechanisms behind PTOA development following ACL injury, we profiled ACL injury-induced gene expression changes in knee joints of three mouse strains with varying susceptibility to PTOA: STR/ort (highly susceptible), C57BL/6 (moderately susceptible) and super-healer MRL/MpJ (not susceptible) and identified genes differentially expressed between these strains at 0-day [before injury], 1-day, 1-week, and 2-weeks post-injury. This study highlights many new potential therapeutic targets and OA biomarkers.

Project description:Synovium is acutely affected following joint trauma and contributes to post-traumatic osteoarthritis (PTOA) progression. Little is known about discrete cell types and molecular mechanisms in PTOA synovium. We aimed to describe synovial cell populations and their dynamics in PTOA. Single-cell RNA-seq profiling of synovium revealed fibroblasts (comprised of various functional subsets), macrophages, endothelial cells and pericytes to be the predominanent synovial cell types, along with rarer populations including T cells, Schwann cells and mast cells. Cell types undergo profound changes in abundance and gene expression following joint injury.

Project description:Post-traumatic Osteoarthritis (PTOA) occurs in about half of individuals who sustained articular injuries including tibial plateau fracture, meniscus tear, or an anterior cruciate ligament (ACL) rupture. We utilize a noninvasive tibial compression injury (TCI) model that can closely replicate the anterior cruciate ligament rupture in patients, which reliably initiates PTOA development. In other studies we utilize single-cell RNA sequencing to temporally explore differences in immune cell infiltration following injury. Although extreme care is taken during immune cell isolation to prevent contamination from bone marrow, we also wanted to rule out the possibility of contamination from bone marrow immune cells in our knee joint samples. Therefore, we proceeded to compare the neutrophil populations from uninjured joint (joint) to bone marrow (marrow) to identify the similarities and differences between these populations.

Project description:Organs of all animals reliably grow to a specific size and it is widely thought that such developmental growth control is instructed by the Hippo signaling pathway. This is based on studies in flies and mice showing that deregulation of Hippo signaling causes dramatic tissue overgrowth of various organs. Here we use two major developmental paradigms and provide evidence that removing the key transcriptional downstream effectors of the Hippo pathway does not abolish organ growth control. Specifically, eliminating the function of the Hippo downstream co-activators Yki as well as Yap/Taz that complex with TEAD family transcription factors did not interfere with organ growth control in the developing mouse liver or Drosophila eye. Rather, mutant cells were able to proliferate appropriately during development, properly exit the cell cycle, and form organs of normal size. Remarkably, a complementing whole transcriptome analysis of mutant Drosophila eye imaginal discs corroborated these genetic findings. It showed that loss of the single TEAD transcription factor Scalloped (Sd) fully reverted the effect of overexpression of hyperactivated Yki back to a wildtype gene expression profile. In contrast, Sd/Yap/Taz proved to be crucial for cell survival in injury response models, namely X-ray irradiation of flies and toxic liver injury in mice. Therefore, we propose that Hippo signaling is dispensable for instructing normal organ growth and instead suggest that the classical Hippo mutant overgrowth phenotypes represent a de facto Yap/Taz/Yki gain-of-function situation that does not reflect an endogenous role in growth control. Instead, we hypothesize that the potency of Yap/Taz/Yki to drive tissue overgrowth draws from their broad and primary function in injury responses.

Project description:The Hippo pathway is an emerging signaling cascade involved in the regulation of organ size control. It consists of evolutionally conserved protein kinases that are sequentially phosphorylated and activated. The active Hippo pathway subsequently phosphorylates a transcription coactivator, YAP, which precludes its nuclear localization and transcriptional activation. Identification of transcriptional targets of YAP in diverse cellular contexts is therefore critical to the understanding of the molecular mechanisms in which the Hippo pathway restricts tissue growth. We used microarrays to profile the gene expression patterns upon acute siRNA knockdown of Hippo pathway components in multiple mammalian cell lines and identified a set of genes representing immediate transcriptional targets of the Hippo/Yap signaling pathway. Three mammalian cell lines (HEK293T, HepG2, HaCaT) were transfected with scramble siRNA controls or siRNAs against NF2 and LATS2, two core components of the Hippo pathway, simultaneously. Total RNAs were harvested four days after transfection to reveal the gene expression pattern unsing microarry. YAP and TAZ siRNAs were also transfected along with NF2 and LATS2 siRNAs to identify YAP/TAZ-dependent transcriptional targets upon loss of NF2/LATS2.

Project description:Latency reversing agents (LRAs) such as protein kinase C (PKC) modulators can reduce rebound-competent HIV reservoirs in small animal models. Furthermore, administration of natural killer (NK) cells following LRA treatment improves this reservoir reduction. It is currently unknown why the combination of a PKC modulator and NK cells is so potent and whether exposure to PKC modulators may augment NK cell function in some way. Here, we compare the transcriptional profiles of PKC-treated NK and CD4+ T cells to assess their generalized cellular effects. Transcriptomic profiles from both cell types displayed signatures of cellular activation and enrichment of genes associated with the NFκB pathway. However, the transcriptomic effects of PKC stimulation on NK cells are much milder than that in CD4+ T cells. This finding, combined with our experimental data testing killing capacity of PKC-stimulated NK cells against K562 cells and infected CD4+ T cells suggests that their contribution in “kick and kill” strategies is primarily due to upregulating HIV expression in CD4+ T cells, rather than directly enhancing the effector functions of NK cells. This suggests that PKC modulators are primarily augmenting the “kick” rather than the “kill” arm of this HIV cure approach.
