DNA methylation profiling of PBMCs from 42 esophageal squamous cell carcinoma patients and 30 controls using the Illumina MethylationEPIC array

ABSTRACT: Esophageal squamous cell carcinoma (ESCC) constitutes over 80% of esophageal cancer cases in China, with advanced-stage patients exhibiting a 5-year survival rate below 30%. Early diagnosis is critical for improving survival outcomes. Current clinical diagnostic methods present significant limitations. This study aimed to identify potential non-invasive biomarkers through epigenomic analysis of the DNA methylome in peripheral blood mononuclear cells (PBMCs) from ESCC patients. Using the Infinium MethylationEPIC (935K) array, we profiled genome-wide DNA methylation patterns in PBMCs from 42 ESCC patients and 30 controls. We discovered an episignature of 742 differentially methylated CpGs (DMCs) that effectively discriminates ESCC patients from controls.
