Induction and maturation of human Cerebroids, a unified 3D system with the full cellular diversity of the brain

ABSTRACT: Brain organoids offer unprecedented opportunities to model developmental or pathological CNS processes. However, most existing models lack essential cellular components, limiting their utility for studying brain physiology and disease. Here, we introduce Cerebroids (CBRs), a standalone, self-organising system that endogenously gives rise to all four major brain cell types: neurons, astrocytes, oligodendrocytes, and microglia, while being amenable to the formation of autologous vasculature, accompanied by scRNAseq data to confirm the cellular component fidelity to primary brain tissue.
