CUT & RUN Analysis in Satellite Cells 7 days after Cardiotoxin Injury of Skeletal Muscle

ABSTRACT: The aim of this project is to assess the open chromatin regions, as well as the androgen (AR) and the glucocorticoid (GR) targets in satellite cells 7 days after cardiotoxin injury. Satellite cells were FACS-isolated and then lysed with Triton X-100 and released nuclei were bound to concanavalin-A magnetic beads. Nuclei/bead complexes were incubated with antibodies directed against AR, GR, a histone mark H3K4me2 and an IgG. After wash, nuclei/bead complexes were incubated with A-micrococcal nuclease and chromatin cleavage was initiated with CaCl2.
