Chromatin priming of skeletal stem progenitor cells governs cell fate during bone repair

ABSTRACT: Bone regeneration is driven by skeletal stem/progenitor cells (SSPCs), which form bone and cartilage after injury. SSPCs represent a diversity of cell populations localized in several bone compartments and adjacent skeletal muscles, with different contributions to bone healing. By generating 10X multiome datasets (i.e., combined single-nucleus RNA-seq and ATAC-seq) and CUT&Tag datasets, we uncovered specificing epigenetic priming of SSPCs at steady-state in muscle and periosteum that dictates their fate decision after a bone fracture.
