Comprehensive analysis of the effects of mitomycin C on human conjunctival fibroblasts

ABSTRACT: RNA-Seq analysis was conducted to investigate the effects of mitomycin C (MMC) on human conjunctival fibroblasts (HConFs). Significant changes in gene expression were observed at 3 hours, 3 days, and 14 days after MMC treatment, with 322, 4,231, and 5,095 genes significantly altered compared to controls, respectively. Enrichment analysis revealed a notable upregulation of genes involved in apoptosis and the p53 pathway, as well as a significant downregulation of genes associated with cell proliferation such as those involved in the G2M checkpoint and E2F targets. Furthermore, the expression of genes related to the senescence-associated secretory phenotype, including FGF2, CCL2, and CCL7, was also increased, suggesting that MMC induces cellular senescence in HConFs as indicated by RNA-Seq analysis.
