Single-cell RNA-Seq analysis of rabbit filtering bleb conjunctiva

ABSTRACT: To investigate changes in fibroblasts in tissues after glaucoma filtration surgery (GFS) using mitomycin C (MMC), we performed scRNA-Seq on bleb-conjunctival tissues after GFS using MMC. We successfully sequenced 4349, 3683, 2588, and 2921 single cells from rabbit GFS tissue of control (without surgery), 3 hours, 3 days, and 14 days after surgery, respectively. An unbiased clustering of single cells projected onto a UMAP space identified 21 major clusters. On days 3 and 14 after surgery, the number of identified fibroblasts was reduced to less than 30% compared to the control group, suggesting the effect of MMC on inducing cell death in fibroblasts. Thus, SASP-related factors such as CDKN1A, IL6, CXCL8, CCL2, and CCL7 were upregulated in fibroblasts after surgery.
