Accessible chromatin profile of early life and adult origin B cell populations

ABSTRACT: The adult B cell pool is a developmental mosaic comprising cells of early life (ELO) and adult origin. To further understdand the differences between B cells of each origin we used a genetic time stamping approach to lable ELO B cells and FACS isolate from the spleen naive follicular and marginal zone B cells, B1 B cells from the peritoneal cavity, and memory B cells (MBC) from the peyer's patch. The assay for transposase accessible chromatin sequencing (ATAC-seq) was performed on each population to identify epigenetic programming that corresponded to ELO versus adult derived B cells.

Project description:The adult B cell pool is a developmental mosaic comprising short-lived naïve B cells and long-lived memory. Using genetic time-stamping, we previously showed that early life origin (ELO) B cells contributed substantially to the adult mouse immune system. In this study, we found that ELO B cells share a memory-like signature. Notably, B-1 cells exhibited an enrichment for the PD-L2/CD80 double positive (DP) immunophenotype associated with highly differentiated memory. Indeed, microbial antigen exposure in neonates expanded distinct specificities within the DP B-1 cell compartment, identifying the latter as a reservoir of IgM memory. B cell chronic lymphocytic leukemia (CLL) is a disease marked by the accumulation of memory-like B cells. By applying time-stamping to a mouse model for unmutated CLL (U-CLL), we demonstrated that leukemic expansion is driven by B-1 cell clones arising prior to postnatal day 10. Importantly, B-1 cells in mice and man closely mirrored the molecular profile of U-CLL. These results suggest that an age-associated leukemia can originate from ELO B cells.

Diurnal changes in bacterial settlement on the Peyer’s patch and surrounding mucosa in the rat ileum and its effect against the intestinal immune system

Project description:To investigate how stimulation from indigenous bacteria for a day affects the local transcriptome of both Peyer's patch and surrounding ordinary mucosa in the rat ileum, we treated rats with a nonabsorbable antibiotics cocktail (vancomycin, polymyxin B and sucrose) for antibiotics-treatment group or sucrose solution for control group in drinking water 1 day before euthanasia. We then performed gene expression profiling analysis using data obtained from RNA-seq of ileal Peyer's patch and surrounding ordinary tissue from rats with or without antibiotics treatment.
