Identification and enrichment of human retinal organoid-derived red/green cone-competent precursors with enhanced axon dynamics

ABSTRACT: Cell replacement therapies aimed at restoring central vision require a source of red/green (long/medium wavelength, or L/M) cone photoreceptors with integrative properties. Toward this end, we developed a means to identify early L/M cone precursors from human pluripotent stem cell-derived retinal organoids (ROs) by linking tdTomato to the endogenous THRB2 promoter. Subsequent bulk RNA sequencing of fluorescence-activated cell sorted ROs showed that very early L/M cones were specifically enriched in genes associated with neural development, axon extension guidance, and cell migration.
