RNA-Seq analysis of 2C genes in reporter mESC lines with or without pladienolide B treatment.


ABSTRACT: Zygotic genome activation (ZGA) in early embryos is driven by extensive chromatin remodeling initiated by maternal factors, enabling the activation of cleavage-stage gene programs that underpin totipotency. In mouse embryonic stem cells (mESCs), a similar 2-cell-like (2C) state can be induced experimentally through the expression of specific maternal regulators such as NELFA. The RNA-seq dataset here was used to estimate the impact of pladienolide B treatment in 3 different reporter mESC lines: 1 with NELFA reporter, 1 with NELFA-MERVL deletion and 1 with total NELFA KO.
