Dysregulated Cholesterol Crystal-STING Pathway in Microglia Drives Neuroinflammation after Ischemic Stroke

ABSTRACT: Dysregulated lipid metabolism is increasingly implicated in microglial activation, yet the specific consequences of massive cholesterol overload following permanent ischemia remain elusive. Using single-cell RNA sequencing in permanent distal middle cerebral artery occlusion (dMCAO) and perioperative ischemic stroke (PIS) models, we elucidated the pathogenic role of intracellular cholesterol crystals. We identified a progressive accumulation of cholesterol crystals within peri-infarct microglia during the subacute phase, driven by a specific downregulation of the lipid-processing enzyme lipoprotein lipase (LPL).
