PD-1 Deficiency Exacerbates Mycobacterium abscessus Lung Infection via Metabolic Rewiring and Dysregulated Neutrophil Responses

ABSTRACT: A PD-1-deficient MAB-infected mouse model was established. The impact of PD-1on bacterial clearance, immune cell activity, and metabolism was evaluated via colony counting, histopathology, flow cytometry, multiplex immunofluorescence, transcriptomics, and metabolic profiling. PD-1−/− mice exhibited increased susceptibility to MAB. Late infection featured further Th1 decline, neutrophil rise, and enhanced glycolysis. Peripheral blood displayed lower CXCL9 but elevated IL-6, IL-21, IL-27, CXCL2, CXCL10, and S100A8 production.
