Translatome profiling by parallel RNA-seq and Ribo-seq during DUX-induced reprogramming of mouse embryonic stem cells to a totipotent-like state.

ABSTRACT: This dataset contains parallel RNA sequencing (RNA-seq) and ribosome profiling (Ribo-seq) data from mouse embryonic stem cells (mESCs) undergoing DUX-induced reprogramming toward a 2-cell-like cell (2CLC) state. Samples were collected at three critical time points: 0 hours, 12 hours, and 24 hours post-induction. The data capture the dynamic landscape of both transcript abundance and genome-wide translational activity throughout this cell fate transition. The dataset enables analysis of: 1) Transcriptional changes and translation efficiency (TE) calculations; 2) Identification of genes regulated specifically at the translational level; 3) The relationship between the in vitro reprogramming translatome and in vivo embryonic development stages. Our analyses revealed a profound but transient suppression of global translation during early reprogramming, coupled with a selective upregulation of mitochondrial genome-encoded transcripts at the translational level. These data provide a comprehensive resource for studying translational control mechanisms in cellular reprogramming and early embryogenesis.
