High-Dose Vitamin D3 Supplementation Associates with Innate Immune Chromatin Remodeling and Context-Dependent Transcriptomic Responses in Multiple Sclerosis [ATAC-seq]


ABSTRACT: We performed longitudinal RNA-seq and ATAC-seq profiling of PBMCs from multiple sclerosis patients receiving vitamin D₃ supplementation for day 0 and day 84. MS samples remained molecularly distinct from controls, with supplementation modulating discrete, context-dependent immune pathways rather than inducing global transcriptomic shifts. Integrated analysis identified candidate vitamin D–responsive regulatory mechanisms with potential relevance to MS pathophysiology.
