Coordinating catalytic and non-canonical functions of EZH2 sensitizes tumors to CAR-T cell therapy

ABSTRACT: This study investigates the dual roles of EZH2 (catalytic and non-canonical) in regulating tumor cell adhesion and sensitivity to CAR-T cell therapy. We performed RNA-seq and CUT&Tag-seq (anti-EZH2) on three human triple-negative breast cancer cell lines (MDA-MB-436, MDA-MB-468, HCC1806) treated with the EZH2 inhibitor EPZ-6438. Additionally, RNA-seq was performed on MDA-MB-468 cells with EZH2 knockdown to dissect the EZH2-dependent transcriptomic underlying tumor cell adhesion and CAR-T therapy response.
