Knockdown of endothelial Serpine1 improves stroke recovery by attenuating peri-infarct blood flow and blood brain barrier disruption

ABSTRACT: This study investigated how endothelial Serpine1/PAI-1 signalling influences cerebral blood flow and functional recovery after stroke. Contrary to expectations, endothelial Serpine1 knockdown reduced peri-infarct capillary flow yet improved blood-brain barrier integrity, reduced inflammation, and enhanced functional recovery, suggesting that carefully regulating — rather than maximizing — post-stroke blood flow may promote recovery.
