Characterization of Human Umbilical Cord Plasma MicroRNAs at Birth and Their Association with Birth-Related Biological Factors, Maternal and Fetal Exposomes, and Factors Related to Early-Life Adiposity Programming

ABSTRACT: To characterize the human umbilical cord plasma (UCP) microtranscriptome profile at birth, we sequenced microRNAs in UCP samples collected at delivery, between the 34th and the 42th weeks of pregnancy (mean gestationnal age at birth: 39.3 ± 1.3 weeks), from 286 newborns from the Gen3G cohort. We then performed association analyses to assess the microRNA profile correlation with birth-related biological factors, maternal and fetal exposomes as well as factors related to early-life adiposity programming.

Project description:Preterm birth, defined as birth <37 weeks of gestation, is a leading cause of infant morbidity and mortality. In the United States, approximately 12% of all births are preterm.1 Despite decades of research, there has been little progress in developing effective interventions to prevent preterm birth. In fact, the rate of preterm birth has increased slightly over the last several decades.2 The ultimate goal of the Genomic and Proteomic Network for Preterm Birth Research (GPN-PBR) is to identify possible biomarkers that could predict the susceptibility to spontaneous preterm birth (SPTB) as well as to shed light on the molecular mechanisms involved in its etiologies. Understanding those mechanisms will help us predict SPTB and may facilitate the introduction of more effective prevention and treatment strategies.

Project description:Using RNA-sequencing (RNA-Seq and miRNA-Seq), we analyzed paired villous trophoblast and decidual basalis transcriptomes of 15 women pregnant with singleton gestations grouped as follows: (1) spontaneous preterm birth (PTB) in the setting of amniocentesis-proven intra-amniotic infection (IAI) and histological chorioamnionits (n=5; GA median [range]: 26 [25-31] weeks); (2) spontaneous idiopathic preterm birth (iPTB, n=5, GA: 32 [30-33] weeks); and (3) term normal pregnancy, that delivered a heathy infant by cesarean section in the absence of labor (n=5; GA: 39 [38-39] weeks).

Liverpool Preterm Birth Biomarker Study

Project description:Preterm birth is a multifactorial condition defined as birth less than 37 weeks of gestation. This study aimed to identify early pregnancy biomarkers from maternal genome-wide and transcriptome-wide data of women experiencing two types of spontaneous PTB (sPTB): spontaneous preterm birth (SPTB) and preterm premature rupture of membranes (PPROM). Whole blood samples were obtained from women with singleton pregnancies (N=567) recruited at the Liverpool WomenÃ¢Â€Â™s Hospital at 16 and 20 weeks of gestation. Women with a previous history of sPTB pregnancies <34 weeks were categorised on their index pregnancy outcome as spontaneous preterm birth (SPTB) and preterm premature rupture of membranes (PPROM) or high-risk term control (HTERM, Ã¢Â‰Â¥37 weeks). Women with no history of SPTB/PPROM and delivered at term were LTERM (Ã¢Â‰Â¥39 weeks of gestation). DNA was genotyped on the UK Biobank AxiomÃ¢Â„Â¢ array (Applied BiosystemsÃ¢Â„Â¢, Thermo Fisher Scientific) and RNA who processed using the ClariomÃ¢Â„Â¢ D Human assay (Thermo Fisher Scientific). Genome-wide association analysis, differential expression analysis and expression quantitative trait loci (eQTL) were completed.

Multi-omics Approach Reveals Metabolic Changes in the Heart at Birth

Project description:During late gestation, the fetal heart relies primarily on glucose and lactate to support rapid growth and development. While numerous studies describe changes in heart metabolism a few weeks after birth to preferentially utilize fatty acids, little is known about metabolic changes of the heart within the first day of life. Therefore, we used the ovine model of pregnancy to investigate metabolic differences between the near-term fetal and the newborn heart. We observed greater abundance of metabolites involved in butanoate and propanoate metabolism, and glycolysis/gluconeogenesis in the term fetal heart (FDR-corrected p<0.10) and differential expression in these pathways were confirmed with single-sample gene set enrichment analysis (ssGSEA) (FDR-corrected p<0.05). Immediately following birth, newborn hearts displayed enrichment in purine, fatty acid, and glycerophospholipid metabolic pathways, as well as oxidative phosphorylation with significant alterations in lipids and metabolites to support transcriptomic findings. While other studies suggest a switch from carbohydrate metabolism to fatty acid metabolism in the neonatal heart in as early as 2 weeks following birth, our data show that this metabolic switch in the heart begins by the first day of postnatal life. A better understanding of metabolic alterations that occur in the heart following birth may improve treatment of neonates at risk for heart failure.
