ABSTRACT: CRISPR Screen with single-cell RNA-Seq as readout reveals new epigenetic factors regulate pluripotency of m ESCs. This highly sensitive method identified key genes essential for the acquisition and exit from pluripotency, and revealed loss of Nsd1-mediated H3K36me2 delayed exit from the naive state, while loss of Ezh2-mediated H3K27me3 promoted transition to the primed state.
