Direct RNA sequencing of in vitro transcribed eGFP mRNA modified with 5-methoxyuracil

ABSTRACT: The aim of this study was to understand potential biases in the incorporation of modified nucleotides, specifically 5-methoxyuracil, in in vitro transcription reactions by different T7 polymerases through Nanopore Direct RNA Sequencing. Incorporation by a range of T7 polymerases using different DNA templates was sequenced using RNA004 flow cell chemistry. Analysis was performed using basecall error-based modification detection (EpiNano).
