Bulk RNA sequencing of whole-blood leukocytes from specific pathogen-free and IMPACT4-exposed mice

ABSTRACT: Specific pathogen-free laboratory mice exhibit limited immune maturation because of restricted microbial exposure. To determine how exposure to a defined consortium of enteric microbes alters systemic transcriptional programs, whole blood was collected from specific pathogen-free control mice and mice exposed to the IMPACT4 microbial consortium. RNA was extracted from the blood leukocytes and subjected to bulk RNA sequencing. Gene-expression profiles were compared between the two experimental groups to characterize transcriptional programs associated with microbial exposure and systemic immune maturation.
