Chaperone condensates buffer the heat shock response against pleiotropic inputs [RNA-seq]


ABSTRACT: RNA-seq of Saccharomyces cerevisiae profiling the heat shock response (HSR) across chemically distinct stresses and upon acute nuclear depletion of the co-chaperone Sis1. Wild-type cells were profiled unstressed and under heat shock, sodium arsenite, azetidine-2-carboxylic acid (AZC), diazaborine (DZA), and hyperosmotic NaCl, each with and without cycloheximide (CHX) to test translation-dependence. In parallel, a rapamycin-resistant Sis1 anchor-away strain was profiled with and without rapamycin across the same stresses. Only heat shock and, weakly, arsenite induce HSR target genes; nuclear Sis1 buffers HSR output under sub-saturating stress.
