Transcriptomic signatures induced by single versus dual lipoparticle-formulated adjuvants

ABSTRACT: Adjuvant delivery using lipid-polymer hybrid nanoparticles (Lipoparticles) represents a promising strategy to modulate immune cell activation. This study investigates whether the combination of two adjuvants encapsulated within Lipoparticles induces distinct transcriptional responses compared with individual adjuvant formulations. Bone marrow-derived macrophages (BMDMs) generated from five independent mice were treated with single or dual adjuvant formulations, and RNA sequencing was performed to characterize genome-wide gene expression changes. Differential gene expression and functional enrichment analyses were conducted to identify biological pathways modulated by these formulations, with a particular focus on immune-related responses.
