Transcriptional consequences of viral and cellular ORF overexpression in hTERT-RPE1 cells


ABSTRACT: To define the transcriptional programs driven by individual viral and cellular open reading frames, hTERT-RPE1 cells were stably transduced with lentiviral constructs expressing GFP11 (negative control), the human papillomavirus E7 oncoprotein, the Epstein-Barr virus protein BNLF2b, or the cAMP-dependent protein kinase inhibitor alpha (PKIA). After puromycin selection, total RNA was profiled by 3' end counting RNA-seq in four biological replicates per condition. Comparison of each ORF-expressing population against the GFP11 control defines the gene expression changes attributable to that ORF.
