Transcriptomic profiling of NCI-H441 epithelial cells following BCG exposure in a human cell-line lung-on-chip model

ABSTRACT: This study investigated early transcriptional responses of NCI-H441 lung epithelial cells in a human cell-line lung-on-chip (CLLOC) model following epithelial-side exposure to Bacillus Calmette-Guérin (BCG). The CLLOC model contained NCI-H441 epithelial cells in the epithelial compartment and human umbilical vein endothelial cells (HUVECs) in the vascular compartment, with human monocyte-derived macrophages incorporated into the epithelial side. BCG was applied to the epithelial/apical compartment, while phosphate-buffered saline (PBS) was used as the control. NCI-H441 epithelial cells were collected from CLLOCs at 12 h post-stimulation for bulk RNA sequencing.
