Single-nucleus transcriptomics reveals coordinated neuronal, immune, and glial dysregulation in the dorsolateral prefrontal cortex in major depressive disorder

ABSTRACT: We used single-nucleus RNA sequencing to define cell-type-specific molecular alterations in the dorsolateral prefrontal cortex (dlPFC) of individuals with MDD. We assessed cell-type composition, differential expression, pathway enrichment, oligodendrocyte-lineage pseudotime, ligand-receptor communication, transcription-factor regulon activity, and overlap with MDD-linked genetic variants. The integrated atlas resolved excitatory and inhibitory neuronal subclasses, oligodendrocyte-lineage cells, astrocytes, microglia/perivascular macrophages, endothelial cells, and vascular-associated populations.
