Identification of osteopontin as a positional and functional candidate gene for cardiac hypertrophy in the SHRSP rat


ABSTRACT: Left ventricular mass is a highly heritable, polygenic trait and linkage studies in WKY and SHRSP rats identified a quantitative trait locus for left ventricular mass index on chromosome 14. Congenic strains, where trait-associated genetic loci are introduced into “healthy backgrounds, identify causal mediators relevant to human disease. Chromosome 14 congenic (WKY.SPGla14a), WKY, and SHRSP strains underwent cardiac phenotyping and transcriptome profiling at; 1-3 days (neonate), 5 weeks, and 16-weeks). Gene expression was assessed in the whole heart of 1-day neonate rats, and the LV of 5 and 16 week rats (N=3/strain) using the RatRef-12 v1.0 microarray chip (Illumina). Whole hearts from 1-day neonate animals were collected, weighted and snap frozen for RNA extraction. LV was snap frozen following dissection and weighting from 5 and 16 week rats. RNA was extracted from frozen heart/LV tissue using RNeasy kits according to the manufacturer’s protocol (Qiagen). Biotinylated amplified target cDNA was prepared and hybridised to the chip.
