Decoding Immune Cell Subset Crosstalk in Biomaterial-Mediated Bone Regeneration: A Sequential Two-Stage Neutrophil-Macrophage Circuit


ABSTRACT: This study uses single-cell RNA sequencing to profile the early immune cell landscape during biomaterial-mediated bone regeneration in a rat calvarial defect model treated with a Janus microsphere delivery system. We identify a sequential two-stage immune circuit wherein CXCL2+ macrophages recruit pro-inflammatory N1 neutrophils via CXCL2-CXCR2 to initiate inflammation, while Neu2 neutrophils subsequently drive macrophage M2 polarization through Lgals9-CD44 to resolve inflammation and initiate osteogenesis-angiogenesis coupling. These findings reveal novel immune subset crosstalk mechanisms and provide a theoretical basis for designing immunomodulatory bone repair materials.
