Transcriptomic profiling of LPS-stimulated RAW 264.7 macrophages with or without osteopontin treatment

ABSTRACT: Osteopontin (OPN/SPP1) has been implicated in the persistence of inflammatory macrophages in osteoarthritis synovium. This study examined transcriptomic changes induced by OPN in LPS-stimulated RAW 264.7 mouse macrophages. Cells were treated with LPS alone or pretreated with recombinant mouse OPN before LPS stimulation, and RNA-seq was performed using three independent biological replicates per condition. The resulting profiles were used to evaluate OPN-associated transcriptional programs in inflammatory macrophages, including pathways related to cell proliferation and apoptosis.
