Liver transcriptome of ANDY mice on niacin-free and control diets compared with aging in wild-type mice (RNA-seq)

ABSTRACT: RNA-sequencing of liver tissue from ANDY mice (transgenic mice with doxycycline-inducible expression of human ACMSD, conferring acquired dependency on dietary niacin for NAD synthesis) and from wild-type C57BL/6J mice, used to compare transcriptional changes associated with chronic NAD deficiency to those associated with natural aging. Male ANDY mice received a niacin-replete control diet (CD) or niacin-free diet (NFD), with doxycycline induction, for 6 months (or, for one short-term arm, 10 weeks); additional arms in the same sequencing batch included a control-diet group that did not receive doxycycline (no transgene induction) and a group on a second niacin-deficient diet formulation. Young (3-month-old) and aged (28-32-month-old) wild-type mice were included for comparison with natural aging. Liver was collected for bulk RNA-seq to characterize transcriptional changes associated with NAD deficiency and aging (Fig. 6a-d).
